清瘟败毒饮及其拆方组对ALI大鼠的干预作用

摘要
目的:研究清瘟败毒饮及其拆方组对大肠杆菌内毒素(lipopolysaccharide,LPS)诱导的大鼠急性肺损伤(acute lung injury,ALI)的干预作用。方法:70只SD大鼠随机分为对照组、模型组、阳性组、清热泻火组(C1)、清热凉血组(C2)、清热燥湿组(C3)和其他组(C4),每组10只,气管内滴注LPS建立大鼠ALI模型。观察多形核白细胞(PMNs)数量、蛋白浓度、肺的湿/干比值、肿瘤坏死因子-α(TNF-α)及肺组织的病理形态学变化。结果:清瘟败毒饮拆方组C1、C2、C3的PMNs及白细胞总数较LPS组显著降低(P<0.05),且C3组最佳(P<0.01);4个拆方组能明显降低肺的湿/干比值(P<0.01),尤其C3组最为显著;C1、C2与C3组明显降低BALF中蛋白浓度(P<0.05),尤其C2组最强(P<0.01);拆方组C1、C2和C4显著降低TNF-α水平(P<0.01),尤其C1组最佳。同时,各拆方组均能降低肺间质及肺泡充血、水肿程度以及炎性细胞的浸润情况。结论:清瘟败毒饮清热泻火和清热凉血拆方组均能较好地改善大鼠ALI损伤,综合各指标情况,可以选择对其拆方组进行进一步药效物质基础研究。
        Objective: To investigate the therapeutic effect of Qingwen Baidu Yin and its separated herbal groups on acute lung injury( ALI) induced by lipopolysaccharide( LPS) in rats. Methods: 70 SD rats were randomly divided into 7 groups,i. e. the blank control group,the model group,the positive control group,C1 group( herbal group of clearing heat to purge fire),C2 group( herbal group of clearing heat to cool blood),C3 group( herbal group of clearing heat and toxic materials),and C4 group( the rest herbs),with 10 rats in each group. The ALI model was established by intratracheal administration of lipopolysaccharide. The percentage of polymorphonuclear leukocytes( PMNs),WBC count,protein concentration,wet/dry ratio of lungs,TNF-α,and pathomorphological changes of lungs were observed 12 h after LPS administration. Results: PMNs and WBC were significantly decreased in C1 group,C2 group,and C3 group,compared to the model group( P <0. 05),and C3 group decreased most( P < 0. 01). All separated herbal groups significantly decreased lung wet/dry ratio( P < 0. 01),of which the effect of C3 group was most obvious. Protein concentrations in BALF were significantly decreased in the groups of C1,C2 and C3( P < 0. 05),of which C2 group had the strongest effect( P < 0. 01). Levels of TNF-α were significantly reduced in groups of C1,C2 and C4( P < 0. 01),of which the effect of C1 group was strongest. Meanwhile,congestion and edema of pulmonary interstitium and pulmonary alveoliall,and inflammatory cell infiltration were all alleviated in the separated herbal groups. Conclusion: C1 group and C2 group can inhibit the inflammatory reaction of ALI and reduce the inflammatory injury to protect the lungs. Therefore,groups of C1 and C2 separated from Qingwen Baidu Yin can be selected for in-depth study.
