补中益气丸对脾虚大鼠能量物质及AMPK的影响

摘要
目的:从ATP、AMP、AMPK的变化探讨脾虚证与能量代谢的关系。方法:采用高效液相色谱法检测大黄脾虚大鼠ATP、AMP的变化,荧光定量PCR方法检测AMPK-α基因的表达。结果:脾虚模型血浆、肝组织AMP/ATP均增高;肌肉中AMPK-α基因表达显著降低,肝组织中AMPK-α基因表达显著升高;补中益气丸对上述模型中异常的能量物质有调节作用。结论:脾虚静息状态下ATP、AMP及AMPK在不同组织中表达不同,肝组织能量需求大于肌肉组织,补中益气丸可以调整脾虚状态下紊乱的能量物质。
        Objective: To discuss the relationship of spleen deficiency syndrome and energy metabolism based on the changes of ATP,AMP and AMPK. Methods: Spleen deficiency syndrome rat model was made by intragastric administration of Rheum. ATP and AMP in plasma,muscle and liver were detected by HPLC. AMPK-α expression in muscle and liver were detected by RT-PCR. Results: Compared with normal group,AMP / ATP ratio were increased obviously in the plasma and liver of model group. AMPK-α gene expression in muscle of model decreased obviously but increased in liver. Buzhong Yiqi pill could regulate the abnormal energy substance above in the model group. Conclusions: There are some differences of ATP,AMP and AMPK in different tissues,and energy supply in liver could be more than in muscle in the spleen deficiency syndrome. Buzhong Yiqi pill could regulate the disordered energy substances in the spleen deficiency syndrome.
