摘要
目的研究彝良小草坝乌天麻提取物对东莨菪碱所致记忆障碍模型小鼠的影响,为阐明天麻道地性及临床应用价值提供实验依据。方法采用腹腔注射东莨菪碱建立小鼠记忆障碍模型,将ICR小鼠随机分为6组:空白组、模型组、石杉碱甲组、小草坝乌天麻水提物低、中、高剂量组。通过Morris水迷宫实验观察彝良小草坝乌天麻对小鼠记忆的影响差异,并测定海马、皮层组织的乙酰胆碱酯酶(acetylcholinesterase,AchE)、超氧化物歧化酶(superoxide dismutase,SOD)、丙二醛(malondialdehyde,MDA)的活性。结果小草坝天麻水提物能缩短东莨菪碱所致记忆获得障碍模型小鼠的逃避潜伏期及搜索距离(P<0.05),明显增加目标象限时间百分比(P<0.01),提高小鼠脑组织中的乙酰胆碱含量(P<0.01)和SOD活力(P<0.05),降低小鼠脑组织中的MDA含量(P<0.01)。结论小草坝天麻水提物能明显改善模型小鼠的记忆获得能力,其作用与增强胆碱能神经系统功能及增强抗氧化能力有关。
        Aim To study the effects of the extract of Gastrodia elata Bl. from Yiliang on mice with memory impairment induced by scopolamine and to provide experimental basis for clarifying the genuineness and the clinical value of Gastrodia elata Bl..Methods The method of intraperitoneal injection of scopolamine was used to establish a mouse model of memory impairment. ICR mice were randomly divided into six groups: blank control group, model group, positive drug group, Gastrodia elata Bl low dose group, middle dose group, and high dose group. Morris water maze was used to examine the influences of Gastrodia elata Bl. from Yiliang on the memory, and the levels of AchE, SOD and MDA in the hippocampus and cortex were determined as well.Results The total extract of Gastrodia elata Bl. from Yiliang could shorten the escape latency and distance in target quadrant of scopolamine-induced memory impairment model mice(P<0.05), significantly increase the original quadrant time percentage(P<0.01), increase acetylcholine content(P<0.01) and SOD activity(P<0.05) in brain tissues of mice, and decrease MDA content(P<0.01) in brain tissues of mice.Conclusions The total extract of Gastrodia elata Bl. from Yiliang can obviously improve the memory acquisition ability of the model mice, which is related to the enhancement of the function of the cholinergic nervous system and thus the antioxidant capacity.
