老年大鼠股动脉结扎加股动静脉吻合与单纯股动脉结扎的侧支血管生长情况的差异性分析

摘要
目的观察老年大鼠单纯股动脉结扎和股动脉结扎加股动静脉吻合对侧支血管生长情况的影响。方法选取36只健康的老年SD大鼠,根据随机数字表法将其分为假手术组(12只)、单纯结扎组(12只)和结扎吻合组(12只)。假手术组大鼠不行股动脉结扎和股动静脉吻合,单纯结扎组大鼠行单纯股动脉结扎,结扎吻合组大鼠行股动脉结扎加股动静脉吻合。所有大鼠手术后存活7d,每组取6只大鼠通过明胶四氧化三铅行血管造影后进行X线拍照对其后肢侧支血管生长情况进行观察,剩下6只取后肢带侧支血管肌肉做冰冻切片,采用免疫荧光组织化学法对Ki67、eNOS及Ang-1进行测定。比较三组大鼠后肢侧支血管数量、Ki67、eNOS及Ang-1免疫荧光强度。结果三组大鼠后肢侧支血管数量、Ki67、eNOS及Ang-1免疫荧光强度存在显著差异(P<0.05);单纯结扎组和结扎吻合组大鼠后肢侧支血管数量、Ki67、eNOS及Ang-1免疫荧光强度显著高于假手术组,差异有统计学意义(P<0.05);结扎吻合组大鼠后肢侧支血管数量、Ki67、eNOS及Ang-1免疫荧光强度显著高于单纯结扎组,差异有统计学意义(P<0.05)。结论血流切应力的提高能够有效促进老年大鼠后肢侧支血管生长,在血管生长过程中,Ki67、eNOS及Ang-1在血管表达增加。
        Objective To observe the effect of simple femoral artery ligation and femoral artery ligation plus femoral arteriovenous anastomosis on collateral vessel growth in aged rats.Methods Thirty-six healthy aged SD rats were divided into Sham operation group(n=12),simple ligation group(n=12)and ligation anastomosis group(n=12)according to random digital table method.In sham-operation group,femoral artery ligation and femoral arteriovenous anastomosis were not performed.Simple femoral artery ligation was performed in the simple ligation group,and femoral artery ligation plus femoral arteriovenous anastomosis was performed in the ligation and anastomosis group.All the rats survived 7 days after operation.Six rats in each group were taken by gelatin trilead tetroxide to take X-ray photographs to observe the vascular growth of the lateral branches of their hind limbs.The Ki67,eNOS and Ang-1 were detected by immunofluorescence histochemistry.The number of collateral vessels and the immunofluorescence intensity of Ki67,eNOS and Ang-1 were compared among three groups.Results There were significant differences in the number of collateral vessels,the immunofluorescence intensity of Ki67,eNOS and Ang-1 among the three groups(P<0.05).The number of collateral vessels,the immunofluorescence intensity of Ki67,eNOS and Ang-1 in ligation anastomosis group and simple ligation group were significantly higher than that in Sham operation group,the difference was statistically significant(P<0.05).The number of collateral vessels,the immunofluorescence intensity of Ki67,eNOS and Ang-1 in the ligation and anastomosis group were significantly higher than those in the simple ligation group,the difference was statistically significant(P<0.05).Conclusion The increase of blood flow shear stress can effectively promote the growth of collateral vessels in the hindlimb of aged rats.During the process of vascular growth,the expression of Ki67,eNOS and Ang-1 increases in the blood vessels.
