摘要
目的:观察健脑通胶囊对AD大鼠一氧化氮合酶(NOS)含量的影响。方法:84只SD大鼠,随机挑出12只,作为空白组,剩余72只给予AD模型复制,造模成功后死亡3只,剩余69只随机分成5组:AD模型组(12只)、脑复康组(12只)、健脑通胶囊小剂量组(15只)、健脑通胶囊中剂量组(15只)、健脑通胶囊大剂量组(15只)。健脑通胶囊大、中、小剂量组分别以剂量0.94g·kg-1·d-1、0.47g·kg-1·d-1、0.235g·kg-1·d-1灌胃;脑复康组予以50mg/mL的脑复康片混悬液按0.162g·kg-1·d-1的剂量灌胃,空白组、模型组予以等量生理盐水灌胃,每天上午给药1次,连续给药30d。测定各组大鼠脑组织NOS含量。结果:健脑通胶囊各组与脑复康组NOS含量均高于模型组(P<0.05);且健脑通胶囊大、中、小剂量组间比较,差异均有统计学意义(P<0.05,P<0.01)。结论:健脑通胶囊能减少AD模型大鼠NOS含量,延缓老年痴呆的发展,有效保护脑功能,且有一定的量效关系。
Objective: To observe the effect of Jiannaotong capsules on nitric oxide synthase( NOS) content in rats with Alzheimer's disease( AD). Methods: Eighty-four Sprague-Dawley rats were included in the study. Twelve of the rats were randoml group,and 72 of them were treated to establish an AD model. Three rats died after the model was established. The other 69 rats with AD were randomly divided into AD model group( n = 12),piracetam group( n = 12),and low-,middle-,and high-dose Jiannaotong capsule groups( n = 15 for each). The high-,middle-,and low-dose Jiannaotong capsule groups were intragastrically given Jiannaotong capsules at 0. 94,0. 47,and 0. 235g·kg-1·d-1,respectively; the piracetam group received suspension of piracetam tablets( 50mg/ml) at 0. 162g·kg-1·d-1; the blank group and model group received an equal volume of normal saline by gavage. Medication was given once daily in the morning for 30 d. Determination of NOS content in brain tissue in rats of each group. Results: All Jiannaotong capsule groups,as well as piracetam group,had significantly higher NOS than the model group( P < 0. 05); there were significant differences between high-,middle-,and low-dose Jiannaotong capsule groups( P < 0. 05 or P < 0. 01). Conclusion: Jiannaotong capsules can decrease NOS content,delay disease progression,effectively protect brain function in AD rats,and has a certain dose effect recationship.
