健脾灸对佐剂性关节炎大鼠心肌超微结构、RLX、ET及NO的影响

摘要
目的:观察健脾灸对佐剂性关节炎(adjuvant arthritis,AA)大鼠心肌超微结构的影响及AA大鼠松弛素(relaxin,RLX)和内皮素-1(endothelin,ET-1)及一氧化氮(nitric oxide,NO)水平的影响。方法:将52只Wistar雄性大鼠随机分为空白组、模型组、药物组和艾灸组,每组13只。各组大鼠右后足内注射弗氏完全佐剂(Freund′s complete adjuvant,CFA)致炎,复制成AA模型。分别在造模前1 d、致炎后每3天、治疗第20天检测4组大鼠体质量、足跖肿胀度及关节炎指数(arthritis index,AI)。治疗20 d后,采用电镜观察大鼠心肌超微结构的变化,同时采用实时荧光定量PCR、Western blot法检测AA模型大鼠RLX和ET-NO系统各指标改变情况。结果:与空白组比较,其他3组大鼠心肌超微结构均有损伤,其中模型组较为明显。与模型组比较,艾灸组、药物组大鼠治疗后RLX、NO水平显著降低,ET-1显著升高(P<0.05或P<0.01);足跖肿胀度、关节炎指数显著降低(P<0.01)。结论:健脾灸可以改善AA大鼠心肌超微结构的损伤,降低关节炎指数、足跖肿胀度,降低RLX、NO水平,升高ET-1水平。
        Objective:To observe the effects of Jianpi Moxibustion on myocardial ultrastructure,relaxin(RLX),endothelin(ET) and nitric oxide(NO) levels in rats with adjuvant arthritis(AA).Methods:52 Wistar male rats were randomly divided into blank group,model control group,drug treatment group and moxibustion treatment group,with 13 rats in each group.Rats in each group were injected with Freund′s complete adjuvant(CFA) into right hind foot to induce inflammation,and then duplicated into AA model.Body mass,foot swelling and arthritis index(AI) were measured 1 day before modeling,3 days after inflammation and 20 days after treatment.After 20 days of treatment,the changes of myocardial ultrastructure in rats were observed by electron microscopy,and the changes of RLX and ET-NO system in AA model rats were detected by real-time fluorescence quantitative PCR and Western blot.Results:Compared with the blank group,all the other three groups had myocardial ultrastructure damage,especially in the model group.Compared with the model group,the levels of RLX and NO in the moxibustion group and drug group were significantly lower.ET-1 was significantly higher(P<0.05 or P<0.01),and the swelling degree of the plantar and arthritis index were significantly lower(P<0.01).Conclusion:Jianpi Moxibustion can improve myocardial ultrastructure damage,reduce arthritis index,foot swelling degree,reduce RLX and NO levels,and increase ET-1 level in AA rats.
