胃癌组织中ADD1、Ptch1、Gli1表达观察

摘要
目的观察胃癌组织中细胞膜骨架蛋白α-Adducin(ADD1)及Hedgehog(Hh)信号通路分子跨膜蛋白Patched1(Ptch1)、转录因子Gli1的表达变化,并分析其相关性。方法行手术治疗的胃癌患者60例,术中留取胃癌组织和癌旁正常胃组织各60例。采用Western blotting法检测胃癌组织及正常胃组织中的ADD1、Ptch1、Gli1蛋白,采用real-time PCR法检测ADD1、Ptch1、Gli1 mRNA。分析ADD1、Ptch1、Gli1蛋白及mRNA表达与胃癌临床病理参数的关系,分析胃癌组织中ADD1、Ptch1、Gli1蛋白及mRNA表达的相关性。结果胃癌组织中ADD1、Ptch1、Gli1蛋白及mRNA相对表达量均高于正常胃组织(P均<0.05)。低分化、Ⅲ~Ⅳ期、有浆膜浸润、有淋巴结转移的胃癌组织中ADD1、Ptch1、Gli1蛋白及mRNA表达分别高于高分化、Ⅰ~Ⅱ期、无浆膜浸润、无淋巴结转移的胃癌组织(P均<0.05)。胃癌组织中ADD1蛋白与Ptch1蛋白、Gli1蛋白表达均呈正相关关系(r分别为0.759、0.681,P均<0.01),ADD1 mRNA与Ptch1 mRNA、Gli1 mRNA表达也呈正相关关系(r分别为0.811、0.719,P均<0.01)。结论胃癌组织中ADD1、Ptch1、Gli1蛋白及mRNA呈高表达,ADD1、Ptch1、Gli1可能共同参与胃癌的发生发展,且ADD1与Hh信号通路之间可能存在一定相互作用关系。
