姜黄素对小鼠急性酒精肝损伤的保护作用

摘要
目的:研究姜黄素减轻酒精导致急性肝损伤的保护功能。方法:60只小鼠被随机均分为5组,分别为空白对照组、乙醇模型组、治疗低剂量组、中剂量组、高剂量组。每日经口灌胃给予受试样品,空白对照组和模型对照组给予蒸馏水。给予受试样品结束时将乙醇模型组及各治疗组一次灌胃给予50%乙醇,空白对照组给蒸馏水。检测小鼠肝脏匀浆液的丙二醛(MDA)、谷胱甘肽(GSH)、甘油三酯(TG)以及血清中丙氨酸氨基转移酶(ALT)和天门冬酸氨基转移酶(AST)。结果:与模型组比较,各治疗剂量组肝匀浆液中MDA和TG水平(MDA:17.237±2.214,12.511±3.791,17.377±4.641;TG:0.446±0.075,0.512±0.132,0.479±0.136)(P<0.05)均显著降低,GSH显著升高(GSH:40.918±8.324,20.787±12.606,21.377±12.041)(P<0.01);血清AST及ALT水平(AST:173.650±12.607,110.879±10.945,127.399±8.212;ALT:65.988±13.588,45.957±9.304,55.745±15.802)均显著降低(P<0.01)。结论:姜黄素能够减轻酒精诱导的急性肝损伤。
        Aim: To study the protective effect of curcumin to alleviate acute liver injury caused by alcohol. Method: 60 mice were randomly divided into 5 groups: blank control group,ethanol model group,the curcumin treatment low dose group,middle dose group and high dose group(40,80,120 mg/kg BW). Mice in the treatment groups were intragastrically infused with different doses of curcumin daily,and the blank control group and model control group were given distilled water daily. At the end of the test,the ethanol model group and each treatment group were given 50% ethanol at one time and the distilled water was given to the blank control group. The levels of malondialdehyde(MDA),glutathione(GSH),triglyceride(TG)in the liver homogenate of mice were estimated. The levels of serum alanine aminotransferase(ALT)and aspartate aminotransferase(AST)were measured. Results: Compared with the ethanol model group,from low dose treatment to high dose treatment,the levels of MDA and TG in the liver homogenate were significantly decreased(MDA: 17.237±2.214,12.511±3.791,17.377±4.641;TG: 0.446±0.075,0.512±0.132,0.479±0.136)(P<0.05),and the levels of GSH was significantly increased(GSH: 40.918±8.324,20.787±12.606,21.377±12.041)(P <0.01). The levels of serum AST and ALT were significantly decreased(AST: 173.650±12.607,110.879±10.945,127.399 ±8.212;ALT: 65.988 ±13.588,45.957±9.304,55.745±15.802)(P<0.01). Conclusion: Curcumin can mitigate the alcohol-induced acute hepatic injury in vivo.
