基于当归/白芍提取物调节原发性痛经大鼠子宫供血的机制研究

摘要
目的探讨当归/白芍提取物调节原发性痛经大鼠子宫供血的作用机制。方法雌性未孕Wistar大鼠随机分为空白组、模型组、治疗组、白芍组、当归组,采用缩宫素建立原发性痛经大鼠模型,用ELISA法检测各组大鼠血清中丙二醛(Malondialdehyde,MDA)、超氧化物歧化酶(Superoxide dismutase,SOD)、精氨酸加压素(Arginine vasopressin,AVP)、缩宫素(Oxytocin,OT)的浓度及大鼠子宫组织中前列腺素F2α(Prostaglandin F2α,PGF2α)、前列腺素E2(Prostaglandin E2,PGE2)、一氧化氮(Nitric oxide,NO)、内皮素(Endothelin,ET)的含量。结果与空白组比较,模型组大鼠血清中MDA、AVP、OT浓度明显升高,SOD浓度降低,子宫组织中PGF2α、PGE2含量均明显升高,NO含量显著下降,ET的含量显著上升(P<0.05)。与模型组比较,治疗组大鼠血清中MDA、AVP、OT浓度明显降低,SOD浓度显著升高,大鼠子宫组织中PGF2α、PGE2含量明显降低,NO含量增加,ET含量明显下降,差异具有统计学意义(P<0.05)。结论当归/白芍提取物能通过抑制子宫收缩,调节血管舒缩物质,改善子宫供血,从而达到对原发性痛经的治疗作用。
        Objective To determine the mechanism of Angelica sinensis(Paeonia lactiflora Pall.) extract in regulating uterine blood supply in rats with primary dysmenorrhea. Methods Female non-pregnant Wistar rats were randomly divided into a control group, a model group, a treatment group, a Radix Paeoniae Alba group, and an Angelica group. Oxytocin was used to establish the primary dysmenorrhea rat model. ELISA was used to detect the serum MDA, SOD, AVP, and PGF2α, and uterine tissue concentration of OT, PGE2, NO and ET. Results Compared with those of the blank group, in the the model group rats the serum MDA, AVP, and OT concentration increased significantly, the concentration of SOD reduced, while PGF2α and PGE2 in uterine tissues was significantly increased. NO content was decreased significantly, and the content of ET was increased(P < 0.05). Compared with those of the model group, the treatment group serum MDA, AVP, and OT concentrations were significantly decreased, SOD concentration increased significantly, PGF2α and PGE2 content in the rat uterus decreased, NO content increased and ET content decreased; with significant difference(P< 0.05). Conclusion Radix Angelica Sinensis and Radix Paeoniae Alba extract can inhibit uterine contraction, regulate vasomotor substances, improve uterine blood supply, which have therapeutic effect on primary dysmenorrhea.
