摘要
目的采用响应曲面法优化知母总黄酮的提取工艺,讨论知母总黄酮的抗氧化活性。方法以提取溶剂、提取时间、提取温度为影响因素,总黄酮得率为评价指标,在单因素试验的基础上采用响应曲面法优化提取工艺。并采用DPPH,ABTS及FRAP三种方法测定提取物的抗氧化活性。此外,利用聚酰胺柱色谱对总黄酮提取物进一步的分离纯化,并测定其抗氧化活性。结果知母总黄酮最佳提取条件为乙醇体积分数48%,提取时间90 min,提取温度100℃,总黄酮提取量2.27 mg/g。经过聚酰胺柱色谱分离,75%乙醇-水洗脱馏分的黄酮含量最高,同时抗氧化活性最好。结论响应曲面法能有效地优化知母总黄酮提取工艺,相关性分析结果表明总黄酮含量与抗氧化活性间存在着显著的正相关性。
        Objective Response surface methodology was employed to optimize conditions for the extraction of total flavonoids from Anemarrhena asphodeloides Bge,and to investigate the antioxidant activities of total flavonoids.Methods The extraction rate of total flavonoids was used as index to evaluate the technology based on single factor test combined with response surface method,in which the extraction solvent,extraction time,extraction temperature were considered as influence factors. DPPH,ABTS and FRAP methods were used to test the antioxidant activities. Polyamide column was applied to purify the flavonoids.Results The best extraction conditions were as follows: ethanol concentration was 48%,extraction time was 90 min and extraction temperature was 100℃. The optimized extraction rate could reach 2. 27 mg/g. It indicated that 75% ethanol-water elution fraction from the polyamide column had the highest content of total flavonoids and with the highest antioxidant activities.Conclusions Response surface methodology can effectively optimize extraction of total flavonoids from Anemarrhena asphodeloides Bge,the concentration of total flavonoids significantly correlated to the on antioxidant activities.
