摘要
随着SSR分子标记技术在玉米室内测纯、一致性及真实性鉴定等方面的应用,该技术已经逐渐成为种子企业质量检测后控的基本手段。但由于其检测量大,准确性要求高的特点,使其对DNA提取质量的要求增加。该文以快速DNA提取方法为基础,在普通的实验条件下,设置4种不同沸水浴的碱煮时间,比较提取DNA后的电泳效果,寻求适合一般种子企业实验室SSR分子试验要求的DNA快速提取方法,为种子企业的快速质量测定奠定基础。
With the use of SSR technology in the aspects of corn chamber test purity,consistency and authenticity identification,this technology has gradually become the seed enterprise quality testing after the control of the basic means.Due to its high detection quantity and high accuracy,the requirement of DNA extraction quality is increased.In this paper,based on the previous human rapid DNA extraction method,under the general experimental conditions of seed enterprises,four different boiling water bath alkali boiling time was set to compare the effect of DNA extraction after electrophoresis,to find a suitable method for rapid DNA extraction in the laboratory of seed enterprises,so as to lay a foundation for rapid quality determination of seed enterprises.
