摘要
目的观察辛伐他汀对矽肺大鼠模型的影响,探讨其治疗矽肺的作用机制。方法 40只SD大鼠随机分为4组:正常对照组(蒸馏水10mL/kg)、矽肺模型组、辛伐他汀小剂量组(10mg/kg)、辛伐他汀大剂量组(20mg/kg),每组10只。采用气管内注入SiO2造成大鼠矽肺模型,连续给药28d后,观察辛伐他汀对矽肺大鼠肺系数、全肺干重/体重比值、肺组织形态学改变,检测SOD活力、MDA、NO含量的变化。结果辛伐他汀可显著降低矽肺大鼠体重、肺系数及全肺干重/体重比值,明显改善矽肺大鼠肺泡炎细胞浸润、充血水肿及肺间质纤维化程度,降低矽结节分级计分,明显提高矽肺大鼠肺组织匀浆总SOD活力,降低总MDA、NO含量。结论辛伐他汀对大鼠矽肺有治疗作用,其作用机制与降低自由基水平有关。
Objective To observe the effects of simvastatin on silicosis in rats and the possible mechanism of the therapeutic effects.Methods Silicosis of rats was induced by intratracheal injection of silicon dioxide.Forty rats were randomly divided into 4 groups: normal control group(distilled water,10 mL·kg-1),model group and simvastatin groups(10 mg/kg and 20 mg/kg),with 10 rats in each group.After the rats were treated for 28 days with corresponding intragastric administration,their lung index,ratio of dry weight of whole lung to body weight,lung histomorphology,SOD activities as well as MDA and NO contents were detected to evaluate the effects of simvastatin on silicosis in rats.Results Simvastatin reduced the body weight,lung index,and ratio of dry weight of whole lung to body weight.Simvastatin ameliorated obviously inflammation cell infiltration,congestion and dropsy of alveoli pulmonis,pulmonary interstitial fibrosis,and decreased the grade scores of silicotic nodule.It increased markedly SOD activities,but decreased MDA and NO contents in lung tissues of silicosis rats.Conclusion The therapeutic effect of simvastatin on silicosis in rats is related to the decreased level of free radicals.
