摘要
目的探讨间充质干细胞对矽尘所致SD大鼠肺损伤的修复作用。方法取8只无特定病原体级健康雄性SD大鼠培养间充质干细胞。另取24只同类大鼠随机分为静脉组、染矽尘组和对照组。静脉组鼠尾静脉注射细胞密度为5×109/L共1m L的间充质干细胞。染矽尘组和染矽尘组气管内注射质量浓度为40 g/L的矽尘混悬液1.0 m L,对照组大鼠气管内注射1.0 m L生理氯化钠溶液。4周后处死大鼠,观察其肺部组织是否病变,同时检测并计算肺脏脏器系数。结果对照组肺脏器系数分别低于染尘组和静脉组(P<0.05);静脉组的肺脏器系数低于染尘组。静脉组BALF总细胞、淋巴细胞及中性粒细胞均低于染尘组(P<0.05)。在病理学检查中,静脉组炎症与纤维化程度与染尘组相比均较轻。在各组血生化指标中,各组血清Cr、CK、ALT、LDH、BUN、AST水平分别比较,差异均无统计学意义(P>0.05)。结论间充质干细胞矽尘所致的肺损伤有一定的修复作用。
        Objective: To investigate the effect of mesenchymal stem cells on the lung injury induced by silica dust in SD rats. Methods: 8 healthy male SD rats with no specific pathogen were cultured. The other 24 rats were randomly divided into two groups. The control group and the vein group were randomly divided into two groups. In intravenous group, the cell density was 5 * 109/L, and the cell density was * 1ml. The silica dust suspension was 1 g/L and the control group was injected with 1 ml, and the control group was injected with 40 ml sodium chloride solution. After 4 weeks, the rats were killed to observe the pathological changes of lung tissue, and to detect and calculate the lung organ coefficient. Results: the lung organ coefficient of control group was lower than that in the group of dust exposed group and vein group(P<0.05). In pathological examination, the degree of inflammation and fibrosis in the vein group were lighter than that in the group of dust. Blood biochemical indicators in each group, the serum Cr, CK, ALT, LDH, BUN, AST levels were compared, the difference was not statistically significant(P>0.05). Conclusion: mesenchymal stem cells have a certain effect on lung injury induced by silica dust.
