摘要
目的:探讨槲皮素对对乙酰氨基酚诱导的人肝细胞LO2的抗氧化作用及可能的凋亡机制。方法:采用碱沉淀酸水解法从槐米提取分离出槲皮素,通过CCK8法测定0-160mM的槲皮素对人正常肝细胞(LO2细胞)增殖的影响,采用Western blot检测槲皮素对APAP诱导LO2细胞损伤模型凋亡蛋白Caspase-9的表达的影响,结果:槲皮素在实验浓度范围内对LO2细胞没有细胞毒性,槲皮素各剂量组的Caspase-9表达呈现下调趋势。结论:槲皮素对APAP氧化损伤的肝细胞具有保护作用,其机制可能与下调的Caspase-9表达有关。
        Objective: To investigate the antioxidant effect and the possible apoptosis mechanism of quercetin on human norma hepatocyte LO2 cells injured by APAP. Methods: Injury model with LO2 cells was established and induced by APAP, and investigated the effect within 160 mM quercetin, the proliferation of the cells was determined by CCK8 assay. The expression of the apoptotic protein caspase-9 was detected by Western blot. Results: Quercetin did not exert cytotoxic effects on LO2 cells in the concentration range of 0-160 m M. Compared with the model group, the expression of caspase-9 in the quercetin group was downregulated. Conclusion: Quercetin has protective effects on APAP oxidatively injured LO2 cells, and its mechanism might be related to down-regulated expression of caspase-9.
