摘要
目的探讨醋延胡索饮片及其颗粒剂HPLC指纹图谱与镇痛药效之间的相关性,即谱-效关系。方法建立醋延胡索饮片及其颗粒剂HPLC指纹图谱,对两者的化学成分组成进行差异性分析。采用热板法和醋酸扭体法评价镇痛药效,采用灰色关联度分析法研究延胡索的谱-效关系。结果建立了不同批次醋延胡索饮片、颗粒剂的HPLC指纹图谱,其中饮片确定了15个共有峰,颗粒剂确定了14个共有峰。醋延胡索饮片及其颗粒剂均能提高小鼠痛阈值,以低剂量颗粒剂痛阈提高百分率最为显著。镇痛谱-效关联度从高到低依次为(峰号) 7> 6> 5> 11> 13> 14> 15> 12> 4> 10> 9> 3> 2> 1,并通过对照品指认了5个相关度较大的色谱峰。结论通过谱-效关系明确了延胡索镇痛作用是由原阿片碱、延胡索甲素、延胡索乙素、小檗碱和巴马汀等多种成分共同作用的结果。
        AIM To study the relationship between the chemical constituents and the analgesic effect of Corydalis pieces and granules. METHODS HPLC fingerprints and chemical compositions of Corydalis pieces and granules was established and analyzed. The analgesic effect of Corydalis pieces and granules was observed by mice hot-board and acetic-acid-induced twisting experiments. The spectrum-effect relationship was studied by gray relational analysis after statistical analysis. RESULTS The HPLC fingerprints of different batches of Corydalis pieces and granules were established,fifteen common peaks were identified in the pieces,and fourteen common peaks were identified in the granules. The analgesic effect results showed that Corydalis pieces and granules could increase the pain threshold,with low dose Corydalis granules increased significantly. The analgesic spectrum-effect correlation result was as following order(peak) 7 > 6 > 5 > 11 > 13 > 14 > 15 > 12 > 4 > 10 > 9 > 3 > 2 > 1,and the top five peaks with strong contribution to analgesic effects were identified. CONCLUSION The analgesic effect of Corydalis pieces and granules is the result of combination of various components,such as protopine,tetrahydropalmatine,corydaline,berberine and palmatine.
