摘要
目的制定笔管草的质量标准,提供其质量控制的依据。方法观察笔管草药材性状和显微鉴别特征,以笔管草对照药材进行笔管草的薄层色谱鉴别。采用高效液相色谱(HPLC)法测定药材中山柰酚的含量。色谱柱为Thermo BETASIL C18;流动相为乙腈-0.4%磷酸溶液(32∶68,V/V);检测波长为365 nm;柱温为30℃;进样量10μL。结果笔管草的性状和显微鉴别特征显著;薄层色谱斑点清晰,分离良好; HPLC法测定笔管草中山柰酚的含量在1.22~381.00μg·m L~(-1)的范围内呈良好的线性关系(Y=40.178 2X+27.948 4,r=0.999 9),测得5批笔管草的山柰酚含量平均值为1.5 mg·g~(-1),加样回收率平均值为94.6%,RSD=3.12%(n=6)。结论所建立的质量标准控制方法简便准确,重现性好,可用于笔管草药材的质量控制。
        OBJECTIVE To establish the quality standard for the Equisetum ramosissimum subsp. debile and provide the basis for its quality control. METHODS The characteristics and microscopic identification of the medicinal herbs were observed. The TLC identification of the herbs was compared with the reference crude herb. Herbs were determined by HPLC method in kaempferiagalanga phenol content: Chromatographic column for Thermo BETASIL C18.Mobile phase of acetonitrile-0.4% phosphoric acid solution( 32 ∶68,V/V).Detection wavelength of 365 nm.The column temperature was 30 ℃.Sample quantity 10 μL. RESULTS The characteristics and microscopic identification of the herbs were remarkable. Thin layer chromatographic spots were clear,separation was good.The Equisetum ramosissimum subsp. debilewas determined by HPLC method in kaempferiagalanga phenol,A good linear relation-ship was obtained within the range of 1.22-381. 00 μg·m L~(-1)( Y = 40.178 2 X+ 27.948 4,r = 0.999 9).The average value of kaempferiagalanga phenol content could be measured in the 5 batchs of Equisetum ramosissimum subsp. debile was 1.5 mg·g~(-1),the average value of the adding standard recoveries was 94.6%,RSD = 3.12%( n = 6). CONCLUSION The quality standard control method is simple and accurate,reproducible,and can be used as the quality control of the Equisetum ramosissimum subsp. debile.
