摘要
以重庆市蚕豆褐斑病病叶为试材,采用组织分离的方法得到5株病原菌,研究了病原菌的培养性状,并对18SrDNA基因和EF-1α基因进行序列分析,采用单因素试验设计研究pH、温度、光照对病原菌生长的影响,以期为蚕豆褐斑病的防治提供参考依据。结果表明:在pH为2~11条件下,病原菌均能生长,最适pH为7;病原菌在5~30℃均能生长,菌丝生长最适为20℃,高于35℃菌丝停止生长;菌丝生长对光照不敏感。
Five strains of pathogens were obtained by using tissue isolation method in Chongqing,and the culture traits of the pathogens and the analysis of 18S rDNA gene and EF-1α gene were studied.The pH,temperature and light on the growth of pathogenic bacteria were studied by single factor experiment,in order to provide reference for the prevention and control of broad bean brown spot disease.The results indicated that the pathogen could grow in pH from 2 to 11,while the optimal pH was 7.The pathogen could grow at 5-30℃,and 20℃ was the most suitable for the mycelial growth.The mycelium stopped growth at 35℃;pathogen was not sensitive to light.
