摘要
目的探讨S100A4基因在慢性肾病(CKD)患者肾组织中表达对肾纤维化损伤的调控作用。方法随机选取我院2016年6月至2018年6月收治的慢性肾脏疾病合并肾纤维化的患者45例为试验组,随机选取同期在我院接受治疗的慢性肾病但未合并肾纤维化的患者45例为对照组。所有患者均进行病例穿刺,均采用免疫组化SP法检测纤维化的肾组织中的S100A4蛋白水平;对肾组织进行Masson染色判断CKD肾纤维化分级(Lee氏分级),并且对肾纤维化程度与S100A4蛋白水平相关性进行分析。结果观察组患者肾纤维化Lee氏分级:II级2例,III级9例,IV级29例,V级5例;观察组患者S100A4蛋白IOD为(3265.25±169.19),对照组患者S100A4蛋白IOD为(6398.84±200.86),观察组低于对照组,对比具有差异性(P<0.05);II级患者IOD值为(4011.13±855.99),III级患者IOD值为(3253.39±364.87),IV级患者IOD值为(2100.37±335.98),III级患者IOD值为(752.99±27.31);对比具有差异性(P <0.05)。结论 CKD患者血浆中S100A4蛋白水平越高表明肾组织纤维化分级越高,与肾纤维化损伤的程度呈明显相关关系,表明S100A4基因在肾纤维化损伤对组织表达调控具有一定作用。
        Objective To investigate the regulation of S100 A4 gene expression in renal tissue of patients with chronic kidney disease(CKD). Methods A total of 45 patients with chronic renal disease and renal fibrosis admitted to our hospital from June 2016 to June 2018 were randomly selected as the experimental group. Forty-five patients with chronic renal disease who were treated in our hospital but did not have renal fibrosis were randomly selected as the control group. All patients underwent case puncture. The levels of S100 A4 protein in fibrotic renal tissues were detected by immunohistochemical SP method. The CKD renal fibrosis grade(Lee's grade) was determined by Masson staining of renal tissue, and the degree of renal fibrosis was determined. Correlation with S100 A4 protein levels was performed.Results The Lee group of renal fibrosis in the observation group: 2 cases of grade II, 9 cases of grade III, 29 cases of grade IV, 5 cases of grade V. The IOD of S100 A4 protein in the observation group was(3263.25±169.19), and the IOD of S100 A4 protein in the control group was( 6398.84±200.86). The observation group was lower than the control group, and the comparison was different(P< 0.05). The IOD value of grade II patients was(4011.13±855.99), the IOD value of grade III patients was(3253.39±364.87), the IOD value of grade IV patients was(2100.37±335.98), and the IOD value of grade III patients was(752.99±27.31). The comparison was different(P< 0.05). Conclusion The higher the level of S100 A4 protein in plasma of CKD patients, the higher the grade of renal fibrosis is related to the degree of renal fibrosis injury,indicating that S100 A4 gene has a certain effect on the regulation of tissue expression in renal fibrosis injury.
