摘要
目的:优化鲜何首乌挥发油的最佳提取工艺,并对挥发油中化学成分进行分析。方法:采用水蒸气蒸馏法提取鲜何首乌挥发油,通过正交试验,研究料液比、盐水饱和度、浸泡时间对鲜何首乌挥发油提取得率的影响,并采用GC-MS对其挥发油中化学成分进行分析。结果:鲜何首乌挥发油的最佳提取工艺为:料液比1∶12,盐水饱和度100%,浸泡时间8h,挥发油得率为0.583%。采用气相色谱-质谱联用技术从鲜何首乌挥发油中分离出28个峰,鉴定了其中的20种成分,占总量的86.03%。主要成分是呋喃甲醛、呋喃甲醇、5-甲基-2-呋喃甲醇、苯酚、1-(2,5-二甲基苯基)乙酮、2-异丙叉基环己酮、3-甲基-2-(2-戊烯基)-2-环戊烯酮。结论 :该研究为何首乌的进一步综合开发利用提供了一定实验依据。
Objective:The research aimed to optimize the extraction process of volatile oil from the fresh Polygonum multiflorum Thunb and analyze the constituents of volatile oil.Methods:The volatile oiI was extracted from fresh Polygonum multiflorum Thunb by water steam distillation method.The effects of the ratio of solid to liquid,and brine saturation,and extraction time on the extraction yield of volatile oil from fresh Polygonum multiflorum Thunbwere studied through orthogonal test.The chemical constituents of volatile oil were analyzed by gaschromatography-mass spectrometry(GC-MS).Result:The optimum technology for extracting the volatile oil from the fresh Polygonum multiflorum Thunbwas as follows:the ratio of solid to liquid 1:12,brine saturation100%,extraction time 8 h,and the yield of volatile oil is 0.583%.Twenty eight peaks were separated from the volatile oil from the fresh Polygonum multiflorum Thunb.Twenty of them were identified,which accounted for 86.03% of the gross.The main compounds were furfural,furfuryl alcohol,5-methyl-2-furancarboxaldehyde,phenol,1-(2,5-dimethylphenyl)Ethanone,2-(1-methylethylidene)-Cyclohexanone,3-methyl-2-(2-pentenyl)-2-Cyclopenten-1-one.Conclusion:This study provided scientific basis for furture comprehensive exploitation and utilization of the Polygonum multiflorum Thunb.
