摘要
目的探讨磁珠与血型抗体偶联过程对抗体活性的影响,为后续利用液相芯片技术检测血型提供实验依据。方法分别将抗D血型抗体直接或者利用抗人Ig G间接与磁珠偶联,再利用磁力架、显微镜观察其与D抗原阳性红细胞的反应情况。结果 Ig M型抗D与磁珠偶联后与D阳性红细胞可反应,但Ig G型抗D与磁珠偶联后与D阳性红细胞不反应;利用抗人Ig G作为桥联间接偶联磁珠的Ig G型抗D不能与D阳性红细胞结合;Ig G型抗D与D阳性红细胞形成的抗原抗体复合物与磁珠可进一步偶联。结论血型抗体与磁珠偶联后会对其活性产生一定影响,尤其是Ig G型血型抗体。
Objective To investigate the effect of coupling of magnetic beads and blood group antibody on antibody activity and provide the experimental basis for the subsequent use of beads-based technology to detect blood type. Methods The antibody against D-blood group was directly or indirectly linked to the magnetic beads,and then the reaction of the D-antigen-positive erythrocytes was observed by using the magnetic frame and microscope. Results The IgM-type anti-D antibody coupled with magnetic beads was able to react with D-positive erythrocytes,while the IgG-type anti-D antibody was not;The IgG-type anti-D antibody indirectly coupled with magnetic beads could not react with D-positive erythrocytes; The antigenantibody complexes of IgG-type anti-D antibody and D-positive erythrocytes could be further coupled with the magnetic beads. Conclusion The coupling of blood group antibody and magnetic beads have certain effects on its activity,especially the IgG type blood group antibody.
