摘要
目的:探究针刺对乳腺增生大鼠乳腺组织中Caspase-8mRNA和Caspase-3mRNA表达的影响。方法:选取40只12周龄的健康雌性未孕SD大鼠(SPF级),将其随机分为正常组、模型组、针刺组与三苯氧胺组,每组10只。造模后进行4周的干预治疗,HE染色后光镜下观察各组大鼠乳腺组织病理状态,PCR法检测各组大鼠乳腺组织Caspase-8mRNA和Caspase-3mRNA的表达情况。结果:HE染色显示模型组较正常组出现乳腺小叶增多、腺泡结构紊乱等组织增生现象,Caspase-8mRNA、Caspase-3mRNA表达水平明显降低(P<0.01);与模型组相比,治疗后的针刺组与三苯氧胺组组织增生的病理变化减轻,Caspase-8mRNA、Caspase-3mRNA表达水平明显增高(P<0.01)。结论:针刺对乳腺增生大鼠乳腺组织中Caspase-8mRNA、Caspase-3mRNA表达有一定影响,可能通过干预凋亡通路对大鼠乳腺增生有调节和治疗作用。
Objective: To explore effects of acupuncture on Caspase-8 mRNA and Caspase-3 mRNA expressions of experimental hyperplasia rats' breast gland. Methods: We chose 40 female 12 weeks aged and non-pregnant SD rats and then randomly divided them into four groups: normal group,model group,acupuncture group,tamoxifen group. Each group had 10 rats. The treatment intervention was 4 weeks after the success of disease model. HE staining was used to observe the pathological changes of breast tissue. Real-time PCR technology was used to observe Caspase-8 mRNA and Caspase-3 mRNA expressions. Result:Compared with the normal group,lobuli mammae of the model group increased significantly. The disorder of acinar structure and tissue hyperplasia phenomenon were obvious. The expressions of Caspase-8 mRNA and Caspase-3 mRNA were significantly decreased( P < 0. 01). After treatment,the acupuncture group and tamoxifen group 's pathological changes of tissue hyperplasia were reduced compared with those of the model group. The expressions of Caspase-8 mRNA and Caspase-3 mRNA were significantly increased( P < 0. 01). Conclusion: Acupuncture can effect expressions of Caspase-8 mRNA and Caspase-3 mRNA in breast glands,regulate and treat hyperplasia of mammary glands in rats by intervening the apoptosis pathway.
