UPLC-MS/MS法分析大鼠血清中二仙汤5种抗骨质疏松成分含量

摘要
建立了超高效液相色谱-电喷雾串联三重四级杆质谱法,对大鼠血清中二仙汤抗骨质疏松成分(仙茅苷、淫羊藿苷、小檗碱、阿魏酸、知母皂苷BII)的含量进行了同时测定。通过大鼠灌胃给药20g/kg二仙汤水提物,得到含药血清,经乙腈沉淀蛋白,浓缩定容后进行色谱分析,外标法定量。该方法对5种抗骨质疏松有效成分的检出限为在0.5～5.0ng/mL,加标平均回收率为90%以上,采用该方法分析含药血清中有效成分含量进行测定,发现含量在0.19～0.80mg/L范围内。
The determined anti-osteoporosis active ingredients are curculigoside,icariin,berberine,ferulic acid and timosaponin BII.Aqueous extracts of Erxian decoction were administrated to rats at an oral dose of 20 mg/kg.Drug-containing serum was treated by acetonitrile protein-precipitation,concentrated and analyzed by UPLC-MS/MS.The detection limits of the five ingredients were in the range of 0.5—5.0 ng/mL.The average spiked recoveries were over 90%.The contents of these active ingredients in drugcontaining serum were in the range of 0.19—0.80 mg/L.
