基于大脑黑质纹状体内多巴胺受体亚型含量与表达观察针刺对大鼠脑卒中痉挛状态的影响

摘要
目的:观察分析针刺对脑卒中痉挛状态(SCA)大鼠大脑黑质纹状体中多巴胺(DA)及其受体亚型(D1、D2)含量与表达的影响,从而探讨针刺对缓解SCA的作用机制。方法:将32只SD大鼠随机分为空白组、假手术组、模型组、针刺组(针刺阳陵泉、曲池),每组8只。运用大脑中动脉线栓法制作SCA模型,造模成功后第1天开始治疗,连续3d。以肌张力评分评价模型和针刺疗效,运用ELISA法分析大鼠大脑黑质中DA及其D1、D2受体的含量,RT-PCR法观察大鼠大脑D1、D2 mRNA的表达。结果:治疗前,与空白组、假手术组比较,模型组、针刺组肌张力评分显著升高(P<0.01);治疗后,与模型组比较,针刺组评分显著降低(P<0.01)。与空白组、假手术组比较,模型组DA及其受体D1、D2 mRNA显著降低(P<0.01),与模型组比较,针刺组DA含量显著升高(P<0.01,P<0.05),DA受体D1 mRNA表达显著增加(P<0.05)。结论:针刺可调节大脑黑质纹状体中DA受体亚型含量与表达,从而对SCA产生良好的治疗效果。
        Objective: To observe and analyze the effect of acupuncture on the the rats with SCA based on the quantity and expression of DA and DA receptor subtypes(D1, D2) in the brain substantia nigra corpus striatum in order to explore the mechanism of acupuncture on alleviating the SCA. Methods: A total of 32 SD rats were randomly divided into control group, sham operation group, model group and acupuncture group that include model with acupuncture Yanglingqun(G30) and Quchi(LI11). There were eight rats in each group. The model of SCA was eatablished by middle cerebral artery occlusion. After successful establish model, rats were treated by acupuncture and continued for 3 days. The muscle tone score was used to evaluate the model and acupuncture efficacy. The levels of DA, D1 and D2 in substantia nigra of rats were analyzed by ELISA. The mRNA expression of D1 and D2 in rats substantia nigra were observed by RT-PCR. Results: Compared with the control group and the sham operation group before the treatment, the muscle tone scores were significantly higher in the model group and acupuncture group(P<0.01). Compared with model group after treatment, the score of acupuncture group was significantly decrease(P<0.01). Compared with the control group and the sham operation group, the mRNA expression of DA, D1 and D2 were significantly decreased in model group(P<0.01). Compared with the model group, the expression of DA in the acupuncture group was significantly increased(P<0.01, P<0.05), the mRNA expression of D1 in the acupuncture group had increased(P<0.05). Conclusion: Acupuncture could regulate the quantity and expression of dopamine receptor subtypes in the substantia nigra of the brain, thus producing a good therapeutic effect on SCA.
