摘要
目的探讨miR-221在甲状腺疾病穿刺细胞活检组织及外周血中的定量表达及意义。方法 51例甲状腺结节患者采用超声引导下粗针穿刺细胞活检法获取标本,术前抽取外周血标本,采用定量PCR法检测组织及血浆中miR-221的表达水平。结果 miR-221在甲状腺良性结节组织中的表达与周围正常组织差异无统计学意义(P>0.05),miR-221在甲状腺癌组织中的表达明显高于穿刺病灶周围正常组织(P<0.05);miR-221在不同甲状腺癌肿瘤病理分期及有无淋巴结转移方面表达差异有统计学意义(P<0.05)。甲状腺癌组外周血miR-221水平显著高于健康对照组和甲状腺良性病变组(P<0.05)。结论 miR-221高表达与甲状腺癌的恶化过程关系密切,可为甲状腺癌术前诊断和个体化治疗提供新的思路。
        Objective To study the expression of miR-221 in biopsy tissue of thyroid disease and peripheral blood.Methods Specimens were obtained by ultrasound guided core-needle in 51 cases of patients diagnosed with thyroid nodules.Peripheral blood samples were collected before operation.Quantitative PCR method was used to detect the expression of miR-221 levels in tissue and plasma.Results The difference of the expression of miR-221 in benign lesions and normal tissue was not statistically significant(P>0.05).In thyroid cancer cases,miR-221 expression level in cancerous tissue were significantly higher than that in normal tissue adjacent to thyroid leisions(P<0.05).There were significant differences in the expression of miR-221 in different pathological stages of thyroid carcinoma and in lymph node metastasis(P<0.05).The level of miR-221 in peripheral blood of patients with thyroid cancer was significantly higher than that in healthy control group and in benign thyroid disease group(P<0.05).Conclusion The high expression of miR-221 is closely related to the progression of thyroid carcinoma.,which can provide novel ideas with preoperative diagnosis and individualized treatment for thyroid cancer.
