摘要
目的观察清达颗粒(QDG)对自发性高血压大鼠(SHR)血压、心脏超声指标及病理学检测的影响。方法将12只16周龄SHR大鼠按随机数字表法分为模型组(SHR组)、清达颗粒组(SHR+QDG组),6只同周龄Wistar大鼠为空白组(Wistar组)。SHR+QDG组给予清达颗粒0.8 g/(kg·d),每次灌药1 mL,连续给药8周,SHR组及Wistar组每天给予等量生理盐水。每4周检测1次血压,直到给药后第8周。在清达颗粒给药结束次日,使用彩色超声心动仪测量并记录各组大鼠左室室间隔厚度、左室内径、左室后壁厚度,并计算左室质量、射血分数(EF)和心肌缩短分数(FS)。HE染色法观察各组大鼠心脏病理学改变,Masson染色观察各组大鼠心肌间质及心肌壁内小动脉周围胶原纤维变化,并计算胶原蛋白容积分数(CVF)和心肌小动脉血管周围胶原面积与管腔面积比值(PVCA)。结果与SHR组比较,SHR+QDG组收缩压、舒张压、平均压均显著下降(P <0.05);SHR+QDG组左室后壁厚度、左室质量、CVF、PVCA较SHR组均显著下降(P <0.05),SHR+QDG组左室室间隔厚度、左室内径与SHR组比较差异无统计学意义(P>0.05)。结论清达颗粒可显著降低SHR血压,改善左心室功能并减轻心肌纤维化,对心肌细胞有保护作用。
        Objective To investigate the effects of Qingda granules(QDG)on blood pressure,echocardiographic parameters and pathological detection in spontaneously hypertensive rats(SHR).Methods Twelve 16-week-old male SHR were randomly divided into model group(SHR group,n =6)and Qingda granule group(SHR+QDG group,n =6),respectively.Six Wistar rats were used as blank control group(Wistar group).The rats in SHR+QDG group were administered with0.8 g/(kg·d)at a dose of 1 mL each time for 8 weeks.The rats in SHR group and the Wistar group were given the same amount of normal saline per day.The blood pressure was measured every 4 weeks until the 8 th week after administration.The left ventricular septal thickness,left ventricular diameter,and left ventricular posterior wall thickness were measured and recorded by color echocardiography,and left ventricular mass,ejection fraction(EF),and myocardial fractional shortening(FS)were calculated on the day after the end of administration of QDG.The pathological changes of the hearts in rats were observed by Hematoxylin and Eosin(HE)staining.The changes of collagen fibers around the myocardial interstitial and small arterioles in myocardial wall of the rats were observed by Masson staining.The collagen volume fraction(CVF)and perivascular collagen area(PVCA)around the myocardial arterioles were calculated.Results Compared with the SHR group,the systolic,diastolic,and mean pressures were significantly decreased in the SHR+QDG group(P <0.05).The left ventricular posterior wall thickness,left ventricular mass,CVF,and PVCA in SHR+QDG group were significantly lower than those in SHR group(P <0.05).There was no significant difference in left ventricular septal thickness and left ventricular diameter between SHR+QDG group and SHR group(P >0.05).Conclusion QDG can significantly reduce blood pressure,improve left ventricular function,alleviate myocardial fibrosis,and protect cardiomyocytes in SHR.
