摘要
目的:应用微电极阵列(MEA)技术研究稳心颗粒对兔心室肌晚钠电流(INaL)的影响。方法:新西兰大白兔50只,随机分成5组,对照组(n=10),心衰组(n=10),心衰+稳心颗粒A组(1 g/L,n=10),心衰+稳心颗粒B组(2 g/L,n=10),心衰+稳心颗粒C组(4 g/L,n=10)。迅速开胸取出心脏进行langendroff灌流,用柔性电极贴附左心室,记录场电位时限(fAPD)等电生理信号的变化。结果:与对照组相比,心衰组兔心室肌fAPD由(347.13±12.41)ms延长至(392.52±13.50)ms,激动传导速度(ECV)由(0.68±0.06)m/s减慢至(0.55±0.08)m/s,心率(HR)由(104±7.23)bpm加快至(126±9.52)bpm(均P<0.05)。与心衰组相比,心衰+稳心颗粒A、B、C组fAPD分别由(392.52±13.50)ms缩短至(369.83±10.72)ms、(348.13±12.56)ms、(331.26±9.37)ms;ECV分别由(0.55±0.08)m/s减慢至(0.45±0.10)m/s、(0.43±0.08)m/s、(0.42±0.07)m/s;HR分别由(126±9.52)bpm减慢至(105±8.42)bpm、(97±6.70)bpm、(94±8.62)bpm(均P<0.05)。但稳心颗粒A、B、C组之间ECV和HR变化无明显差异(P>0.05)。结论:稳心颗粒是一种安全有效的抗心律失常中药,对INaL的抑制作用是其发挥抗心律失常作用的主要机制之一。
        Objective:To explore the effect of Wenxin Granule on the late sodium current(INaL) in rabbit ventricular muscle by microelectrode array(MEA)technology.Methods:Fixty rabbits of either sex were randomly divided into 5 groups:control group(n=10),heart failure group(n=10),heart failure+Wenxin Granule A group(1 g/L,n=10),heart failure+Wenxin Granule B group(2 g/L,n=10),heart failure+Wenxin Granul C group(4 g/L,n=10).The heart was perfused with modified Tyrode′s solution after removed.Flexible electrodes were attached to the left ventricle,and we recorded the changes of electrophysiological signals.Results:In the heart failure groups,fAPD prolonged from(347.13 ±12.41)ms to(389.52 ±13.50)ms,ECV decreased from(0.68±0.06)m/s to(0.55±0.08)m/s,whlie HR increased from(104±7.23)bpm to(126±9.52)bpm compared with the control group′s(above all P<0.05).In the heart failure group+Wenxin Granule A,B,C groups,fAPD shortened from(389.52 ±13.50)ms to(367.83 ±10.72)ms,(348.13 ±12.56)ms and(331.26 ±9.37)ms respectively.ECV decreased from(0.55±0.08)m/s to(0.45±0.10)m/s,(0.43±0.08)m/s and(0.42±0.07)m/s respectively,while HR decreased from(126±9.52)bpm to(105±8.42)bpm,(97±6.70)bpm and(94±8.62)bpm(above all P<0.05).However,there were no significant differences in the change of ECV or HR amongst Wenxin Granule A,B and C groups(P>0.05).Conclusion:Wenxin Granule is a safe and effective antiarrhythmic traditional Chinese medicine,and the inhibition of INaL is one of the main mechanisms of its role in antiarrhythmic effect.
