摘要
目的研究开胃消食合剂对功能性消化不良(FD)大鼠的治疗作用及相关机制。方法将60只雄性Wistar大鼠随机分为空白组、模型组、阳性药组、开胃消食合剂低剂量组、开胃消食合剂中剂量组、开胃消食合剂高剂量组,空白组正常饲养,其余采用饥饱失常和夹尾激怒法构建FD模型。持续造模14 d后,灌胃治疗14 d,实验结束。观察各组大鼠状态,记录体重,用葡聚蓝-2000灌胃来测定大鼠的胃排空率和小肠推进率。采用酶联免疫吸附测定(ELISA)检测血清和十二指肠中胃动素(MTL)和生长抑素(SS)的含量。结果与空白组比较,模型组体重下降(P <0.01);与模型组比较,开胃消食合剂中、高剂量组能增加大鼠体重(P <0.05或P <0.01)。与空白组比较,模型组胃排空率降低(P <0.01);与模型组比较,开胃消食合剂高剂量组能增加胃排空率(P <0.05)。与空白组比较,模型组小肠推进率明显降低(P <0.01);与模型组比较,开胃消食合剂低、中、高剂量组小肠推进率均有所升高,但差异无统计学意义(P> 0.05)。与空白组比较,模型组血清和十二指肠中MTL含量明显降低(P <0.05或P <0.01);与模型组比较,开胃消食合剂高剂量组能增加血清和十二指肠中MTL的含量(P <0.05或P <0.01)。与空白组比较,模型组血清和十二指肠中SS含量明显增加(P <0.05);与模型组比较,开胃消食合剂高剂量组能减少大鼠血清和十二指肠中SS含量(P <0.05)。结论开胃消食合剂具有治疗FD的作用,能够增加胃排空率和小肠推进率,其作用机制与改善胃肠激素SS和MTL的分泌密切相关。
        Objective To study the therapeutic effect of Kaiwei Xiaoshi Mixture on functional dyspepsia(FD) rats and its related mechanism. Methods Sixty male Wistar rats were randomly divided into blank group, model group, positive drug group, low dose of Kaiwei Xiaoshi Mixture group, middle dose of Kaiwei Xiaoshi Mixture group and high dose of Kaiwei Xiaoshi Mixture group. The blank group rats were fed normally. The rest of rats were fed with starvation and satiety disorders and tail-clipping irritation for 14 days to establish FD model. The gavage treatment lasted for 14 days.At the end of the experiment, the mental state of rats in each group was observed and the weight of rats was recorded.The gastric emptying rate and small intestinal propulsion rate of rats in each group were measured by gavage with blue dextran 2000. The contents of motilin(MTL) and somatostatin(SS) in serum and duodenal tissue were detected by enzyme-linked immunosorbent assay(ELISA). Results Compared with the blank group, the body weight of the model group decreased(P < 0.01); compared with the model group, the body weight in the middle and high dose of Kaiwei Xiaoshi Mixture groups increased(P < 0.05 or P < 0.01). Compared with the blank group, the gastric emptying rate of the model group decreased(P < 0.01); compared with the model group, the gastric emptying rate in high dose of Kaiwei Xiaoshi Mixture group increased(P <0.05). Compared with the blank group, the small intestine propulsion rate of the model group was significantly decreased(P < 0.01); compared with the model group, the intestinal propulsion rate in low, middle and high dose of Kaiwei Xiaoshi Mixture groups increased, while there were no statistically significant differences(P > 0.05). Compared with the blank group, the MTL content in the serum and duodenum of the model group was significantly decreased(P < 0.05 or P < 0.01); compared with the model group, the MTL content in serum and duodenum of high dose of Kaiwei Xiaoshi Mixture group increased(P < 0.05 or P <0.01). Compared with the blank group, the SS content in the serum and duodenum of the model group was significantly increased(P < 0.05); compared with the model group, the SS content in serum and duodenum of high dose of Kaiwei Xiaoshi Mixture group reduced(P < 0.05). Conclusion Kaiwei Xiaoshi Mixture has the effect of treating FD and can significantly increase gastric emptying rate and small intestinal propulsion rate, and its mechanism is closely related to the improvement of gastrointestinal hormones SS and MTL secretion.
