摘要
试验对HPLC-ELSD法测定食品中果糖、葡萄糖、蔗糖、麦芽糖和乳糖五种糖的方法进行了较为详细的探讨,采用酰胺柱(250 mm×4.6 mm, 5μm),蒸发光散射检测器,漂移管温度80℃,气体压力40 psi,色谱柱温度40℃,进样体积10μL,流速1 mL/min,流动相为乙腈+丙酮(0.2%氨水)︰水(0.2%氨水)=75︰25 (体积比)。结果表明,在10.5min内,五种糖得到充分分离,试验结果重复性良好,线性良好,相关系数均大于0.99,加标回收率在94.7%~110.0%之间,相对标准偏差(n=6)在0.3%~3.0%之间,结果可靠。
This test was discussed in detail aiming at detecting fructose, glucose, sucrose, maltose, and lactose in food using HPLC-ELSD method. Use amide column(250 mm×4.6 mm, 5 μm), evaporative light-scattering detector, drift tube temperature 80 ℃, gas pressure 40 psi, column temperature 40 ℃, injection volume 10 μL, flow rate 1 mL/min, and the mobile phase was acetonitrile and acetone(0.2% ammonia)︰water(0.2% ammonia)=75︰25(volume ratio). The results show that the time of the separation of five kinds of sugar was only 10.5 min, repeatability of test results had good linear,correlation coefficient was greater than 0.99, the standard addition recovery was between 94.7% and 110.0%, relative standard deviation(n=6) was between 0.3% and 3.0%, and the result was reliable.
