摘要
目的研究蒸制轮叶党参(steamed Codonopsis lanceolata,SCL)对乙醇暴露致急性肝损伤小鼠的保护作用。方法雄性ICR小鼠随机分为6组,即空白对照组、模型组、水飞蓟素组及SCL低、中、高剂量组。除空白对照组外,其余各组建立急性酒精性肝损伤小鼠模型。各组给予相应干预措施。采用比色法测定小鼠血清中谷丙转氨酶(ALT)、谷草转氨酶(AST)、三酰甘油(TG)的水平及肝组织中还原型谷胱甘肽(GSH)和丙二醛(MDA)含量,并观察肝组织病理,进行组织病理学评分。结果与空白对照组比较,模型组小鼠血清ALT、AST水平及急性酒精性肝损伤中TG含量升高,肝脏中的MDA水平上升,GSH活性降低,差异有统计学意义(P<0.05)。与模型组比较,SCL给药组能够明显降低小鼠血清中ALT和AST水平及急性酒精性肝损伤中TG含量,抑制肝脏中上升的MDA水平,增强GSH的活性,差异均有统计学意义(P<0.05)。HE结果显示,模型组有明显的肝损伤,表现为细胞坏死、相邻肝细胞间的脂肪变性和淋巴细胞的渗透,而SCL高剂量组没有观察到严重的脂肪变性,肝脏损伤和肝结构的分裂程度最低。结论 SCL对小鼠急性酒精肝损伤具有保护作用,其可能是通过改善脂质过氧化而发挥作用。
        Objective To study the protective effects of steamed Codonopsis lanceolata( SCL) on alcohol-induced acute liver injury in mice. Methods Male ICR mice were randomly divided into the blank control group,model group,silymarin group,SCL groups with low-,medium-and high-dose. Except the blank control group,mice model of acute liver injury was established by alcohol in the other groups. Each group was received the corresponding intervention. The serum levels of alanine aminotransferase( ALT),aspartate aminotransferase( AST),triglyceride( TG) and the liver contents of reduced glutathione( GSH) and malondialdehyde( MDA) were determined by colorimetry. The pathological changes of liver tissue were observed and the histopathological scores were evaluated. Results Compared with the blank control group,the serum levels of ALT,AST and TG were increased in the model group,and the content of MDA was increased and the activity of GSH was decreased in liver,with statistically significant differences( P < 0. 05). Compared with the model group,the serum levels of ALT,AST and TG were significantly decreased in mice treated with SCL,and the content of MDA was decreased and the activity of GSH was increased in liver,with statistically significant differences( P < 0. 05). The results of HE staining showed that the obvious liver injury was observed in the model group,which performed as the cells necrosis,fatty degeneration of adjacent hepatocytes and infiltration of lymphocytes. But no severe steatosis was observed in the SCL group with high-dose,and the liver injury and the fragmentation degree of liver structure was the slightest. Conclusion Steamed Codonopsis lanceolata shows protective effects on alcoholinduced acute liver injury in mice. Its mechanism may be related to the improvement on lipid peroxidation.
