摘要
目的:基于鹿茸-山药药对补肾壮阳活性,探讨鹿茸-山药药对对肾阳虚大鼠的治疗作用。方法:采用雄性Wistar大鼠90只,随机分为空白对照组、模型组[注射氢化可的松2.5 mg·(100 g)~(-1)]、中药阳性药组(男宝胶囊0.6 g·kg~(-1))、西药阳性药组(丙酸睾酮,10 mg·kg~(-1))、鹿山高剂量组(1.8 g生药·kg~(-1))、鹿山中剂量组(0.9 g生药·kg~(-1))、鹿山低剂量组(0.45 g生药·kg~(-1))、鹿茸组(0.4 g生药·kg~(-1))及山药组(1.4 g生药·kg~(-1)),每组10只,于末次给药后测定各组大鼠体质量、血清中环磷酸腺苷(cAMP)、环磷酸鸟苷(cGMP)、睾酮(T)、卵泡刺激素(FSH)、黄体生成素(LH)、促性腺激素释放激素(GnRH)、肾上腺皮质酮(CORT)水平、前列腺系数、提肛肌+储精囊系数。结果:与空白对照组相比,模型组大鼠体质量与血清中cAMP、cAMP/cGMP、T、FSH、LH、GnRH、CORT水平均显著降低(P<0.05或P<0.01),前列腺系数、提肛肌+储精囊系数均显著下降(P<0.05);与模型组比较,中药阳性药组血清cAMP、cAMP/cGMP、T、FSH水平与前列腺系数均显著升高(P<0.05),西药阳性药组FSH水平、LH水平、提肛肌+储精囊系数显著增加(P<0.05),鹿茸组T、CORT、GnRH水平升高(P<0.05),山药组FSH水平显著提高(P<0.05),鹿山高剂量组cAMP、cAMP/cGMP、FSH、LH水平与前列腺系数、提肛肌+储精囊系数提高(P<0.05),鹿山低剂量组大鼠体质量、LH水平显著升高(P<0.05)。结论:鹿茸-山药药对具有补肾壮阳作用,其机制可能是通过二者的协同作用增加cAMP调节垂体-性腺轴功能提高FSH、LH水平与前列腺系数及提肛肌+储精囊系数实现的。
        Objective:To study the effects of Antler-Chinese yam on kidney yang deficiency rat.Methods:Ninety male rats were randomly divided into blank control group,model control group[injection of hydrocortisone 2.5 mg·(100 g)~(-1)],traditional Chinese medicine control group(0.6 g·kg~(-1)),western medicine control group(testosterone propionate,10 mg·kg~(-1)),high dose group of Antler-Chinese yam(1.8 g raw drug·kg~(-1)),and middle dose group(0.9 g raw drug·kg~(-1)),low dose group(0.45 g raw drug·kg~(-1)),antler group(0.4 g raw drug·kg~(-1))and yam group(1.4 g raw drug·kg~(-1)).The influence of each samples on the body weight,serum hormone level and sex organs of hydrocortisone induced Yang deficiency rats were measured.Results:Compared with the blank group,the body weight,serum levels of cAMP and cAMP/cGMP,T,FSH,LH,GnRH and CORT in the model group decreased significantly(P<0.05 or P<0.01),and the prostate coefficient,levator ani muscle and seminal vesicle coefficient decreased significantly(P<0.05).Compared with the model group,the levels of serum cAMP,cAMP/cGMP,T,FSH and prostate coefficient were significantly increased in the Chinese medicine positive group(P<0.05),FSH,LH,coefficient of levator ani muscle and seminal vesicle were significantly increased in the western medicine positive group(P<0.05),T,CORT,GnRH levels in the pilose antler group were significantly increased(P<0.05),and FSH levels in the yam group were significantly increased(P<0.05).The levels of cAMP,cAMP/cGMP,FSH,LH,coefficient of prostate coefficient,levator ani muscle and seminal vesicle in Antler-Chinese yam high dose group were increased(P<0.05),while the body weight and LH levels in Lushan low dose group were significantly increased(P<0.05).Conclusion:The Antler-Chinese yam has the effect of invigorating kidney yang,which may be the synergistic effect of the two drugs to regulate the pituitary-gonadal axis function by increasing the content of cAMP,and improve the level of FSH,LH and the coefficient of gonadal gland.
