摘要
目的研究蜂眠宁胶囊对肾虚型老年痴呆动物模型的改善作用。方法将雄性Wistar鼠随机分为6组,即空白组、假手术组、模型组、六味地黄丸组和蜂眠宁胶囊大、小剂量组。每天灌胃给予氢化可的松(15mg/kg),两周后于侧脑室注射-淀粉样蛋白(25-35)10 g建立肾虚型老年痴呆动物模型。于手术第2天开始,六味地黄丸和蜂眠宁胶囊大、小剂量组灌胃给药。观察、检测各组大鼠的体重及一般状态、学习记忆能力、自主活动,行为学检测后测定各组大鼠血清皮质酮含量、免疫学指标及脑组织病理学变化。结果模型组大鼠体重增长缓慢,自主活动迟缓,水迷宫学习记忆能力减退,胸腺和脾指数降低,血清皮质酮值降低;假手术组各项指标比模型组轻,表明短时间肾虚对学习记忆有一定影响;蜂眠宁胶囊大、小剂量组动物体重增长、自主活动、水迷宫上台潜伏期、实台象限游程、穿越平台次数、胸腺和脾指数、血清皮质酮值等与模型组比较得到明显改善(<0.05,<0.01),效果具有显著性差异,并优于六味地黄丸组(<0.05)。结论蜂眠宁胶囊对肾虚型老年痴呆症状具有显著改善作用。
        Objective To study the effects of Fengmianning capsule on the senile dementia animal models with deficiency of kidney. Methods Male Wistar rats were randomly divided into normal group,model group,control group,Fengmianning capsule(FMN)high dose group, low dose group and Liuwei Dihuang pill(LWDH)group. To establish the senile dementia,the rats of the model group,FMN groups and LWDH group were treated by orally administered hydrocortison(15 mg/kg/d)for14 d and then intracerebroventricular injection of amyloid-protein(25-35)at the 14 th day,control group was given the same volume of normal saline. From the 15 th day to the end,FMN-treated rats and LWDH-treated rats were orally administered,rats in other groups were given an equivalent amount of distilled water. From the beginning,the following changes including body weight,locomotor activity,learning and memory,serum corticosterone,immunological indexes and the histopathology factors of brain were observed. Results The model rats showed that weight grew slowly,autonomic activity decreased,the ability of learning and memory impaired,thymus index,spleen index decreased,the level of serum corticosterone reduced.After 3 weeks of administration of FMN capsule,all such indexes of rats were significantly improved( <0.05, <0.01),while LWDHtreated rats showed the same improvement effects compared with model group.Conclusion FMN capsule and LWDH pill can also significantly improve senile dementia with deficiency of kidney.
