靖州茯苓三萜类成分对A549细胞中Nrf2信号通路的调控机制研究

摘要
目的研究靖州茯苓三萜类成分体外对A549细胞增殖的影响及对核因子E2相关因子2(Nrf2)、谷胱甘肽巯基转移酶(GST)、人醌NADH脱氢酶1(NQO1)蛋白表达的影响。方法采用噻唑盐比色(MTT)法检测A549细胞的存活率,Western blot法测定Nrf2,GST,NQO1蛋白表达水平。结果茯苓三萜质量浓度大于15μg/mL时,A549细胞存活率开始降低,且质量浓度与存活率呈负相关;质量浓度大于30μg/mL时,A549细胞中Nrf2,GST,NQO1蛋白表达水平显著升高(P <0. 05),且此时质量浓度与蛋白表达水平呈正相关。结论靖州茯苓三萜类成分可通过下调Nrf2信号通路相关蛋白的表达抑制A549细胞的增殖。
        Objective To study the effect of triterpenes from Poria cocos in Jingzhou on the proliferation of A549 cells in vitro and the expression of NF-E2-related factor 2( Nrf2),glutathione S-transferase( GST) and human NAD( P) H dehydrogenase,quinone 1( NQO1)protein. Methods The survival rate of A549 cells was measured by MTT assay, and the expression levels of Nrf2, GST and NQO1 protein were determined by Western blot. Results When the concentration of triterpenes from Poria cocos was more than 15 μg/mL,the survival rate of A549 cells began to decrease, and the concentration of triterpenes was negatively correlated with the survival rate. When the concentration of triterpenes from Poria cocos was more than 30 μg/mL,the expression levels of Nrf2, GST and NQO1 protein in A549 cells increased significantly( P < 0. 05),and the concentration of triterpenes was positively correlated with the expression levels of protein. Conclusion The triterpenes from Poria cocos in Jingzhou can inhibit the proliferation of A549 cells by down-regulating the expression of Nrf2 signaling pathway-related protein.
