摘要
研究荞麦叶大百合总黄酮的乙醇提取工艺及其抗氧化活性,在单因素试验基础上,以乙醇浓度、提取温度、料液比、提取时间为自变量,总黄酮提取率为因变量,运用正交试验优化荞麦叶大百合中总黄酮提取工艺。同时,测定荞麦叶大百合总黄酮对DPPH自由基的清除活性。结果表明:荞麦叶大百合中总黄酮提取的最佳工艺条件为乙醇浓度为70%,提取温度60℃,料液比为1∶10(g/mL)和提取时间为30 min。此条件下,总黄酮提取率达到10.90 mg/g,重复性试验结果表明,此方法稳定可靠,提取率高,适于荞麦叶大百合中总黄酮的提取。荞麦叶大百合总黄酮和V_C对DPPH自由基的半数清除率EC_(50)分别是4.439、18.746μg/mL,由抗氧化试验结果看出,荞麦叶大百合中总黄酮对于DPPH自由基的清除能力优于抗氧化剂V_C,具有较强的抗氧化活性。
        Orthogonal test method was used to optimize the extraction technology of total flavonoids from Cardiocrinum cathayanum. On the basis of single-factor test,the extraction conditions(ethanol concentration,extraction temperature,ratio of water to materials and extraction times)were analyzed and optimized by the orthogonal design. Meanwhile,the scavenging effects of total flavonoids from Cardiocrinum cathayanum on DPPH·were determined. The results showed that the optimum extraction conditions were as follows:ethanol concentration was 70 %,extraction temperature was 60 ℃,ratio of raw material to liquid was 1 ∶ 10(g/mL)and extracting for 30 min. Under the conditions,the yield of total flavonoids was 10.90 mg/g. This method was suitable for the extraction of total flavonoids from Cardiocrinum cathayanum with high extraction rate. The total flavonoids from Cardiocrinum cathayanum and vitamin C had the DPPH radical scavenging ability with EC_(50) of 4.439 μg/mL and 11.297 μg/mL,respectively. The results of in vitro antioxidant test showed that the scavenging effects of total flavonoids extract from Cardiocrinum cathayanum to DPPH· was better than that of vitamin C solution at the same concentration.
