摘要
目的:探索生长素IAA对川牛膝Cyathula officinalis Kuan侧根发育和基因IAA14表达的影响。方法:以道地产区乐山金口河3年生川牛膝的种子为材料,在含有不同浓度IAA的MS培养基中培育得到川牛膝幼苗,对其进行根显微结构分析、侧根参数和基因IAA14表达量的考察。结果:川牛膝侧根的起始部分同大多数双子叶植物一样,侧根原基起始于正对木质部的中柱鞘细胞;IAA在一定范围内对川牛膝侧根的发生具有一定的促进作用,且同时下调基因IAA14表达量;平均侧根密度最大值出现在1.83×10~(-5)mg/mL的IAA施加浓度,此时基因IAA14表达量也最低。结论:川牛膝侧根起始于正对木质部的中柱鞘细胞,IAA通过实时调节侧根发育相关基因IAA14在一定范围内促进川牛膝侧根的发育。
        Objective:To investigate the effects of IAA on the development of lateral roots and IAA14 gene expression of Cyathula officinalis.Methods:The seeds of triennial Cyathula officinalis on geo-authentic habitats in Jinkou River of Leshan city were used as materials.Cyathula officinalis seedlings were cultured in MS medium containing different concentrations of IAA.The root microstructure,lateral root parameters,and IAA14 gene expression level were analysis.Results:The origin of lateral roots of Cyathula officinalis was the same as that of most dicotyledons.The lateral root primordium originated from the pericyclic cells directly opposite the xylem;IAA promoted the occurrence of lateral roots of Cyathula officinalis in a certain range.The IAA14 gene expression was down-regulated at the same time.The maximum number of average lateral root density appeared at an applied concentration of IAA with 1.83×10~(-5)mg/mL,and the expression of IAA14 gene was the lowest.Conclusion:The lateral roots of Cyathula officinalis originate from the pericyclic cells directly opposite the xylem,and IAA promotes the development of lateral roots of Cyathula officinalis in a certain range by real-time regulating the lateral root development-related IAA14 gene.
