摘要
目的:探讨疏利气机、涤痰行瘀方蠲哮汤对哮喘大鼠肺组织CysLTs和CysLTR1 mRNA的影响。方法:将189只SPF级雌性SD大鼠随机分为正常组(A组)、模型组(B组)、孟鲁司特钠组(C组)、地塞米松组(D组)、蠲哮汤低、中、高剂量组(E-G组),卵蛋白氢氧化铝溶液致敏,并长期雾化吸入卵蛋白激发哮喘,A组予0.9%氯化钠溶液雾化代替,分别持续1、2、4周,各时间点处死每组大鼠9只,检测肺组织CysLTs和CysLTR1mRNA。结果:肺组织CysLTs:同时间段,与B组比较,C组、D组、F组和G组CYSLTs表达显著下调(P<0.05)。不同时间段,各药物治疗组第4周CYSLTs表达显著低于本组第2周CYSLTs表达(P<0.05)。肺组织CysLTR1m RNA:同时间段,与B组比较,C组、D组、F组和G组CysLTR1 mRNA相对表达水平均显著下调(P<0.05)。不同时间段,各药物治疗组第4周CysLTR1 mRNA表达低于本组第2周CysLTR1 mRNA表达(P<0.05)。结论:蠲哮汤干预哮喘大鼠肺组织CysLTs和CysLTR1 mRNA有效。蠲哮汤可能是通过影响CysLTR1 mRNA表达干预CysLTR1介导的哮喘CysLTs炎症作用达到防治哮喘的目的。
        Objective: To study the effect of Juanxiao Decoction on mRNA expression of CysLTs and CysLTR1 in the lung tissue of asthmatic rats. Methods: A total of 189 SPF female SD rats were randomly divided into contol group(A group),model group(group B), montelukast group(group C), dexamethasone group(D group), Juanxiao Decoction low dose group(group E), Juanxiao Decoction medium dose group(F group) and Juanxiao Decoction high dose group(G group). Asthma model was established by long-term nebulization ovalbumin sensitized aluminum hydroxide solution. A group received 0.9% sodium chloride solution instead of atomization respectively for 1, 2 and 4 weeks, the end of the week, lung tissue was used to detect CysLTs and CysLTR1 mRNA expression in each 9 asthmatic rats. Results: CysLTs in lung tissue: at the same time, compared with the B group, C group, D group, F group and G group each dose group down-regulated expression of CYSLTs(P<0.05). At different time the expression of CYSLTs in each drug treatment group at fourth weeks was lower than the second weeks(P<0.05). Lung tissue CysLTR1 mRNA: at the same time, compared with B group, C group, D group, F group and G group CysLTR1 mRNA relative expression levels were decreased(P<0.05). Compared with the C group, the expression of CysLTR1 mRNA in the treatment group was lower than that of second weeks CYSLTs at fourth week(P<0.05). Conclusion: Juanxiao Decoction had effect to intervene CysLTs and CysLTR1 mRNA in the lung tissue of asthmatic rats. Juanxiao Decoction may be mediated by CysLTR1 expression intervention effect of CysLTs to achieve the aim of preventing and treating asthma inflammation in asthma by CysLTR1 mRNA.
