摘要
目的探讨不同提取溶剂和提取方法对柴胡地上部分总黄酮提取率的影响。方法以浸膏得率和总黄酮提取率作为考察指标,采用紫外线(UV)法,先以35%、45%、55%、65%、75%和85%乙醇作为溶剂,采用室温浸渍法选取最佳提取溶剂,然后以该溶剂作为提取溶剂,利用恒温水浴振荡器提取法、静态热回流提取法、动态热回流提取法、超声波辅助提取法、微波辅助提取法5种提取方法进行提取。结果 55%的乙醇为最佳提取溶剂,其总黄酮提取率和浸膏得率分别为(1.120±0.043)%、(13.600±0.583)%;以55%乙醇作为提取溶剂,所优选的提取方法为静态热回流提取法,总黄酮提取率和浸膏得率分别为(2.220±0.081)%、(16.160±1.650)%。结论柴胡地上部分总黄酮提取的最优提取溶剂是55%乙醇,最优提取方法是静态热回流提取法。
Objective To explore the effects of extraction solvents and extraction methods on the extraction rate of to-tal flavonoids from the aerial part of bupleurumchinense.MethodsWith the extracting rate and the extraction rate of total fla-vonoids as index,used the method of UV,first in 35%,45%,55%,65%,75% and 85% ethanol as solvent,the selection ofthe best extraction solvent at room temperature by immersion method,The best extraction solvent was selected by room tempera-ture immersion method,and then the solvent was used as the extraction solvent by using thermostatic water bath oscillator ex-traction method,static hot reflux extraction method,dynamic hot reflux extraction method,ultrasonic assisted extraction methodand microwave assisted extraction method.ResultsThe best extraction solvent was 55% ethanol and the extraction rate andextraction rate of total flavone were(1.120±0.043)% and(13.600±0.583)%,respectively. Taking 55% ethanol as the extrac-tion solvent,the preferred extraction method was the static thermal reflux extraction method,with the extraction rate of flavoneand extract yield(2.220±0.081)% and(16.160±1.650)%,respectively.ConclusionThe optimum extraction solvent for theextraction of total flavonoids from the aerial part of bupleurumchinense is 55% ethanol,and the optimal extraction method isstatic hot reflux extraction.
