三黄凝胶对大鼠耳廓痤疮模型血清IFN-β和IL-6的影响

摘要
目的观察三黄凝胶对大鼠耳廓痤疮模型血清白细胞介素6(IL-6)、β干扰素(IFN-β)含量的影响,探讨三黄凝胶治疗痤疮的作用机制。方法将48只6～8周龄Wistar大鼠随机分为空白组、模型组、阳性对照组及三黄凝胶低、中、高剂量组,共6组,每组8只,除空白组,其余各组于大鼠双侧耳廓内导管开口处涂抹100%油酸以制造实验大鼠耳廓痤疮模型,连续抹药3周,肉眼及病理学观察提示造模成功后开始外用药物。空白组、模型组不使用任何药物,三黄凝胶低、中、高剂量组分别涂抹不同剂量的三黄凝胶,阳性对照组涂抹0.025%维A酸乳膏,连续治疗4周后,ELISA法检测各组大鼠血清IL-6、IFN-β的含量。结果各治疗组大鼠血清IL-6、IFN-β的含量均明显低于模型组,差异有统计学意义(P<0.01),三黄凝胶低、中剂量组与阳性对照组比较差异无统计学意义(P>0.05),三黄凝胶高剂量组与阳性对照组比较差异有统计学意义(P<0.05)。结论三黄凝胶治疗痤疮的作用机制可能与抑制和减少IL-6和IFN-β的产生有关。
Objective To observe the effect of Sanhuang gel on serum interleukin-6(IL-6) and interferon-beta(IFN-β) in acne model of rats,and to explore the mechanism of Sanhuang gel in the treatment of acne. Methods Forty-eight Wistar rats aged 6～8 weeks were randomly divided into blank group,model group,positive control group and Sanhuang gel low,middle and high dose groups. As a result,six groups was set up with eight in each.Except blank group,100% oleic acid was applied to the opening of bilateral auricular ducts in rats in the other entire group to create earacne models of the experimental rats.Wiping continuously lasted three weeks. Visual and pathological observations indicated that the external use of the drug began after the model was successfully established.No drugs were used in the blank group and the model group. Different doses of Sanhuang gel were applied to the low,middle,and high dose groups of the Sanhuang gel,and 0.025% Vitamin A acid cream was applied to the positive control group. After continuous treatment for 4 weeks,the ELISA was performed. The serum IL-6 and IFN-β levels were measured in each group.Results The serum levels of IL-6 and IFN-β in the treatment groups were significantly lower than those in the model group(P<0.01). There was no statistical difference between the low-dose group and the positive control group(P>0.05). There was a statistically significant difference between the high dose group and the positive control group(P<0.05). Conclusion Sanhuang gel treatment of acne may be related to the inhibition and reduction of IL-6 and IFN-β production.
