摘要
对测定血清中H2S浓度的方法进行了改进。基于2,6-dansyl azide本身不发荧光,但与H2S作用后显示强烈荧光这一特性,一步法合成2,6-dansyl azide后采用PBS/DM SO体系(pH 7.5)替代PBS/Tween-20体系(pH 7.5)测定内源性H2S。数据分析显示改进的方法具有更低的荧光背景值。应用本方法成功对6只成年健康SD大鼠的血浆H2S水平进行了测定。
        An improved method for measuring H2 S concentration in serum was described. The 2,6-dansyl azide(2,6-DNS-Az) itself does not fluoresce,however,it can emit a strong fluorescence when reacts with H2 S. Based on this characteristic,2,6-DNS-Az was synthesized by one-step method,then endogenous H2 S was determined with PBS buffer/DMSO(pH7. 5) instead of PBS buffer or PBS buffer/Tween-20(pH7. 5). The study indicated that the improved assay method has a better determination effect with lower fluorescence background value. Furthermore,this method was used successfully for measuring endogenous hydrogen sulfide concentration in plasma from 6 healthy adult SD rats.
