Survivin与Caspase-3在胆管细胞癌中的表达及相关性研究

摘要
目的探讨Survivin与Caspase-3在胆管细胞癌中的表达,并探讨二者之间的相关性。方法选择2013年12月~2015年12月在我院手术治疗的胆管细胞癌患者40例(A组)手术标本作为研究对象,同时取癌旁胆管组织30例(B组)。另外选取同期手术的10例正常组织(肝血管瘤旁正常肝内胆管组织或者肝外伤后切除肝组织的正常肝内胆管组织)作为对照(C组)。比较各组Survivin与Caspase-3表达的阳性率与临床病理参数的关系,及Survivin在胆管细胞癌中的表达与Caspase-3的相关性。结果 Survivin与Caspase-3在胆管细胞癌中的阳性率分别为75.0%、40.0%,其中Survivin在A组中的阳性率显著高于B组、C组,差异有统计学意义(P<0.05)。而Caspase-3在A组中的阳性率低于B组、C组,差异有统计学意义(P<0.05)。Survivin、Caspase-3在胆管细胞癌中的表达与分化程度及淋巴结转移有密切关系,差异有统计学意义(P<0.05)。胆管细胞癌中Survivin阳性30例,Caspase-3阳性16例,二者均为阳性7例,二者均为阴性1例,应用相关分析显示,Survivin在胆管细胞癌中的表达与Caspase-3呈负相关(r=-0.723,P<0.05)。结论 Survivin在胆管细胞癌中的阳性表达率较高,但Caspase-3较低,二者呈负相关,考虑可能是Survivin抑制了Caspase-3介导的细胞凋亡,对Survivin与Caspase-3的研究有助于深入了解胆管细胞癌的发生、发展机制。
Objective To investigate the expression of Survivin and Caspase-3 in cholangiocarcinoma and to explore the correlation between the two. Methods 40 cases(group A) with cases of bile duct carcinoma were selected as the study subjects, and 30 cases(group B) were collected from the surgical treatment of cholangiocarcinoma in our hospital during December 2013 to December 2015. In addition, 10 cases of normal liver tissue(normal intrahepatic bile duct tissue or liver injury after liver trauma) were selected as control group(group C). The relationship between the expression of Survivin and in each group, the relationship between the positive rate of Caspase-3 expression and the clinicopathological parameters, and the correlation between the expression of Survivin and the expression of Caspase-3 in cholangiocarcinoma were compared. Results The positive rates of Survivin and Caspase-3 in cholangiocarcinoma were 75.0% and 40.0%, respectively. The positive rate of Survivin in group A was significantly higher than that in group B and group C(P<0.05). And the positive rate of Caspase-3 in group A was lower than that of group B and group C(P<0.05).The expression of Caspase-3 and Survivin was closely related to the degree of differentiation and lymph node metastasis, and the difference was statistically significant(P<0.05). Bile duct cell carcinoma survivin in 30 cases of positive,caspase-3 was positive in 16 cases, both were positive in 7 cases, two were negative in 1 case, the application correlation analysis showed that survivin in bile duct carcinoma and the expression of Caspase-3 was negatively correlated(r=-0.723, P<0.05). Conclusion The positive expression of Survivin in cells of bile duct cancer rate is high, but caspase-3 is low, the second is negatively related, consider possible is by inhibition of caspase-3 apoptosis mediated by Survivin. The research of Survivin and Caspase-3 helps to understanding the development mechanism of cholahgiocarcinoma.
