摘要
设计合成了一种基于香豆素结构的荧光探针ZQT-015并表征了其结构。在PBS(0. 1 mmol/L,pH 6. 86)缓冲液中,探针ZQT-015对酪氨酸酶表现出高度的选择性识别作用和良好的抗干扰能力。探针ZQT-015在526 nm处的荧光强度变化与酪氨酸酶浓度呈现良好的相关性。以左旋多巴和探针ZQT-015为底物分别考察了曲酸对酪氨酸酶的抑制能力,拟合得到的IC50值比较接近,表明ZQT-015适用于酪氨酸酶抑制剂的筛选。
        A fluorescent probe ZQT-015 based on coumarin structure was designed,synthesized and well characterized. In PBS( 0. 1 mmol/L,pH 6. 86) buffer,the probe ZQT-015 showed a high selective recognition of tyrosinase and good anti-interference ability. The fluorescence intensity of the probe ZQT-015 at 526 nm showed a good correlation relationship with the tyrosinase concentration. The inhibitory ability of kojic acid to tyrosinase was investigated using levodopa and probe ZQT-015 as substrates,and the IC50 values obtained by fitting were similar,indicating that ZQT-015 is suitable for the screening of tyrosinase inhibitors.
