摘要
目的:建立黄柏药材中总生物碱的含量测定方法。方法:采用酸性染料比色法,通过对pH值,显色剂用量,测定波长等实验条件进行筛选,寻求最佳测定条件。结果:在pH=5.2,0.03%酸性染料3 mL,测定波长为415 nm时,能准确测定黄柏药材中总生物碱的含量,其精密度、线性、重复性、回收率等均良好。结论:本含量测定方法操作方便,重现性好,准确性好,可以用于黄柏总生物碱的含量的测定。
        Subjective: To establish a method for determination of total alkaloids of Phellodendron medicinal materials. Methods: first, the ultrasonic extraction method, extraction of total alkaloids in Cortex Phellodendri, by acid dye colorimetry, by UV sub light photometer and its absorbance was measured, to calculate the content of total alkaloids in Cortex Phellodendri chinensis powder. The concentration of berberine hydrochloride had good linear relationship with 40~200 μg/mL, and the regression equation was y=0.0023 x-0.0604, R2=0.9996(n=5). Results: the measured content of total alkaloids from cortexphellodendri in 4.31 %. The average recovery was 100.37 %. Conclusion: the content determination method has the advantages of convenient operation, good reproducibility, good accuracy, can be used for the determination of the total alkaloids of phellodendron.
