摘要
目的以模式动物秀丽线虫为模型,对以黄芪等为主要成分的复方汤剂开展抗衰老活性研究。方法先检测原始汤剂对秀丽线虫的寿命延长作用,然后对原始汤剂进行萃取分离获得不同提取部位再检测各部位对秀丽线虫寿命的影响,同时采用薄层色谱法鉴定原始汤剂及其提取部位的代表性化学成分。结果原始复方汤剂及其多糖萃取部位具有延缓秀丽线虫衰老的作用,毛蕊花糖苷、芦丁、黄芪甲苷等化合物是复方汤剂的代表性化学成分。结论本文复方汤剂通过多糖成分发挥其抗衰老作用。
Objective To assess the anti-aging activity of a complex decoction composed primarily of Astragalus radix and other herbs using the model animal Caenorhabditis elegans.Methods The anti-aging activity of the initial decoction was examined by lifespan assay using wild-type C.elegans.The decoction was then extracted into 6 fractions using chemical reagents with different polarities,and the fractions were tested for their effects on C.elegans lifspan.Representative compounds in the initial decoction and its fractions were identified by thin layer chromatography.Results The decoction and its polysaccharide fraction was able to prolong C.elegans lifespan.Acteoside,rutin and astragaloside were among the representative compounds in the decoction.Conclusion The complex decoction may exert its anti-aging activity through its polysaccharides.
