摘要
目的:建立同时测定藏药双花千里光中的绿原酸、芦丁、金丝桃苷、异槲皮苷、烟花苷和紫云英苷6个化学成分含量的HPLC方法。方法:双花千里光药材用70%乙醇-水提取;以乙腈(A)-0.1%甲酸水溶液(B)为流动相,梯度洗脱(0~2 min,0→16%A;2~6 min,16%A→15%A;6~8 min,15%A;8~13 min,15%A→20%A;13~16 min,20%A→25%A;16~19 min,25%A→20%A;19~26 min,20%A→30%A),检测波长为360 nm。结果:所测6个化学成分在所取质量浓度范围内的线性关系均良好,r值都大于0.999 0;精密度、重复性和稳定性良好;平均回收率为96.1%~99.4%,RSD为0.84%~1.8%;6个化学成分绿原酸、芦丁、金丝桃苷、异槲皮苷、烟花苷、紫云英苷的含量测定结果分别为1.75%、1.26%、0.86%、1.20%、0.57%、1.29%。结论:建立的HPLC方法可用于同时测定双花千里光药材中6个化学成分,可为藏药双花千里光药材的质量控制提供一定的参考。
        Objective:To establish an HPLC method for simultaneous determination of six chemical components including chlorogenic acid,rutin,hyperoside,isoquercitrin,kaempferol-3-O-rutinoside,and astragalin in Tibetan medicine Senecio dianthus Franch..Methods:The Senecio dianthus Franch.herbal medicine was extracted with70% ethanol-water.The mobile phase was acetonitrile(A)-0.1% aqueous formic acid(B)in gradient elution(0-2 min,0 → 16%A;2-6 min,16%A → 15%A;6-8 min,15%A;8-13 min,15%A → 20%A;13-16 min,20%A → 25%A;16-19 min,25%A → 20%A;19-26 min,20%A → 30%A).The detection wavelength was 360 nm.Results:The linear relationship of the six chemical components was good,and the r values were all above 0.999 0.Results of precision,repeatability and stability tests were good.The average recovery were 96.1%-99.4%with RSDs of 0.84%-1.8%.The contents of the six chemical components were 1.75%(chlorogenicacid),1.26%(rutin),0.86%(hyperoside),1.20%(isoquercitrin),0.57%(kaempferol-3-O-rutinoside),and 1.29%(astragalin).Conclusion:The established HPLC method can be used to simultaneously determine six chemical components in Senecio dianthus Franch..It can provide reference for the quality control of Tibetan medicine Senecio dianthus Franch..
