芍药汤对溃疡性结肠炎大鼠结肠组织病理变化及免疫功能的影响

摘要
目的观察芍药汤对溃疡性结肠炎(UC)大鼠结肠组织病理变化及免疫功能的影响,探究其对UC大鼠的免疫调节机制。方法采用TNBS诱导制作UC模型。实验大鼠随机分为正常组、模型组、美沙拉嗪组和芍药汤组,各给药组给予相应药物灌胃,连续7 d。观察大鼠一般状况、体质量、结肠组织大体形态、结肠黏膜损伤指数(CMDI)评分,HE染色观察结肠组织病理变化,ELISA检测血清白细胞介素(IL)-1β、肿瘤坏死因子-α(TNF-α)、IL-4含量,免疫组化检测结肠组织IL-1β、TNF-α、IL-4蛋白表达。结果与正常组比较,模型组大鼠体质量显著降低(P<0.01),CMDI评分、血清和结肠组织IL-1β、TNF-α含量和蛋白表达显著升高(P<0.01),IL-4含量和蛋白表达显著降低(P<0.01);与模型组比较,芍药汤组和美沙拉嗪组大鼠体质量显著升高(P<0.05),结肠组织一般形态和病理变化显著改善,CMDI评分、血清和结肠组织IL-1β、TNF-α含量和蛋白表达显著降低(P<0.05,P<0.01),IL-4含量和蛋白表达显著升高(P<0.05,P<0.01)。结论芍药汤能有效改善UC模型大鼠结肠组织一般状况及病理变化,提高机体免疫功能,其机制可能与调节IL-1β、TNF-α及IL-4等细胞因子的含量和蛋白表达相关。
        Objective To observe the effects of Shaoyao Decoction on colonic pathological changes and immune function in rats with ulcerative colitis(UC) induced by TNBS; To explore its immunoregulatory mechanism for UC rats. Methods A model of UC was induced using TNBS. SD rats were randomly divided into normal group, model group, mesalazine group and Shaoyao Decoction group. Each administration group was given relevant medicine for gavage for 7 d. The general condition, body weight change, gross morphology of colon tissue and CMDI score were observed. HE staining was used to detect pathological changes of colon tissue. Serum levels of IL-1β, TNF-α and IL-4 were detected by ELISA. Immunohistochemistry was performed to detect the expressions of IL-1β, TNF-α and IL-4 protein in colon tissue. Results Compared with the normal group group, body weight of the rats in model group significantly decreased(P<0.01). The CMDI score, the contents and protein expressions of IL-1β and TNF-α in serum and colon tissue increased significantly(P<0.01), and the content and protein expression of IL-4 decreased significantly(P<0.01). Compared with the model group, body weight of the rats in Shaoyao Decoction group and mesalazine group significantly increased(P<0.05), and the general morphology and pathological changes of the colon tissue were significantly improved. The CMDI score, the contents and protein expressions of IL-1β and TNF-α in serum and colon tissue decreased significantly(P<0.05, P<0.01), and the content and protein expression of IL-4 increased significantly(P<0.05, P<0.01). Conclusion Shaoyao Decoction can effectively improve the general condition and pathological changes of colon tissue in UC rats, and regulate the immune function of the body. The mechanism may be related to the regulation of the contents and protein expressions of IL-1β, TNF-α and IL-4 and other cytokines.
