摘要
目的观察成年偏颌大鼠左右侧嚼肌浅层细胞的超微结构和肌球蛋白重链Ⅱ型基因表达量与正常成年大鼠的差异。方法青春期雄性SD大鼠20只,随机分为实验组和对照组,每组10只。实验组大鼠左侧嚼肌浅层为实验A组,右侧为实验B组。实验组大鼠配戴引导下颌左偏的矫治器,对照组不戴。大鼠成年时处死,取实验A、B组及对照组大鼠嚼肌浅层细胞,用透射电镜观察细胞超微结构,用实时定量PCR技术检测肌球蛋白重链Ⅱ型基因表达量的差异。结果实验A、B组细胞形态改变,细胞内肌丝排列紊乱,线粒体形态数量改变。实验B组内肌浆网多见。实验A组肌球蛋白重链Ⅱ型基因表达量是对照组的162.9%,是实验B组的128.0%(P<0.01),实验B组肌球蛋白重链Ⅱ型基因表达量是对照组的12.7%(P<0.01)。结论偏颌大鼠左右侧嚼肌浅层细胞的超微结构不同,肌肉的收缩力有显著差异,可能对偏颌的产生有一定作用。
        Objective To investigate the ultrastructural characteristics and myosin heavy chain II expression of superficial masseter muscle(SMM)cells in adult rats with mandibular asymmetry. Methods Twenty adolescent male Sprague-Dawley rats were divided into experimental group and control group of ten. The SMM of left side in experimental group was experimental group A. The contralateral side was experimental group B. The experimental group was fitted with fixed devices that made the mandible shift to the left. The control group did not wear the devices. The animals were sacrificed when they became adult.The SMM of the left and right side of the experimental and control group was dissected and observed under transmission electronic microscope. The expression of myosin heavy chain II was examined by real-time quantitative polymerase chain reaction(qPCR). Results The shape of SMM cells in experimental group A and B changed.The arrangement of myofilaments in the SMM cells of both sides in experimental group was disarranged. The amount and shape of mitochondria in the cells of the experimental group changed. There were a large number of sarcoplasmic reticulum cisternae in the right side of experimental SMM cells.The expression of myosin heavy chain II in the experimental group A was 162.9% of the control group,128.0% of the experimental group B(P<0.01). The experimental group B was 12.7% of the control group(P<0.01). Conclusion The ultrastructural characteristics and strength of SMM cells in left and right side of adult rats with mandibular symmetry were different from the normal adult rats. The SMM may have effect on the mandibular asymmetry.
