摘要
目的:基于"肾藏精起亟"理论观察补肾活血中药对慢性心力衰竭(CHF)大鼠骨桥蛋白(OPN)的影响。方法:60只雄性SD大鼠,随机选取15只为假手术组,剩余45只通过冠脉结扎配合力竭式游泳建立CHF大鼠模型,模型成功后随机分为模型组、西药组、中药组。用药4周后,超声检测大鼠心脏形态变化,免疫组化法测定大鼠心肌及肾脏组织中OPN表达水平,Real-time PCR法检测大鼠心肌及肾脏组织中OPN mRNA表达水平。结果:治疗后,与假手术组比较,模型组大鼠左室内径(LVID)显著增高,射血分数(EF)和短轴缩短率(FS)均显著减低(P<0.05),心、肾组织中OPN蛋白及mRNA表达水平显著升高(P<0.05);与模型组比较,西药组、中药组LVID显著降低,EF、FS显著升高(P<0.05),心、肾组织中OPN蛋白及mRNA表达水平显著降低(P<0.05)。结论:补肾活血中药可下调OPN蛋白及mRNA的表达,改善CHF大鼠心室重构,揭示了"肾藏精起亟"心肾相关的中医病机基础。
        Objective: To observe the effect of Bushen Huoxue Formula on the OPN in chronic heart failure rats based on the theory of ‘the kidney's function of storage the essence of life for accommodation'. Methods: Sixty male SD rats were randomly selected as sham operation group, and the remaining 45 rats were randomly divided into model group, western medicine group and traditional Chinese medicine group after successful model establishment by coronary artery ligation and exhaustive swimming. After 4 weeks of treatment, the heart morphology of rats was detected with ultrasound examination, OPN expression in myocardium and kidney of rats was detected with immunohistochemistry, OPN mRNA expression in myocardium and kidney of rats was detected with Real-time PCR. Results: After treatment, compared with sham operation group, left ventricular internal diameter(LVID) of model group rats were increased significantly, ejection fraction(EF) and fractional shortening(FS) of model group rats were decreased significantly(P<0.05), OPN and OPN mRNA expression in myocardium and kidney of model group rats were increased significantly(P<0.05). Compared with model group, LVID of lisinopril and Bushen Huoxue group rats were decreased significantly, EF and FS of lisinopril and Bushen Huoxue group rats were increased significantly(P<0.05), OPN and OPN mRNA expression in myocardium and kidney of lisinopril and Bushen Huoxue group rats were decreased significantly(P<0.05). Conclusion: Bushen Huoxue Formula can reduce the expression of OPN mRNA and OPN, and improve the ventricular remodeling of chronic heart failure rats, reveal traditional Chinese medicine pathogenesis of ‘kidney's function of storage the essence of life for accommodation' with heart and kidney related theory.
