摘要
目的:考察钴60(~(60)Co-γ)射线辐照灭菌效果及对天花粉有效成分含量的影响。方法:选择辐照剂量为0,5,8,10kGy对天花粉药材辐照灭菌,采用微生物限度检查法考察灭菌前后需氧菌总数和霉菌、酵母菌总数变化; HPLC法比较有效成分葫芦素B、葫芦素E的含量变化,并检验其显著性情况。结果:天花粉经8 kGy辐照剂量灭菌可达到中国药典微生物限度要求;天花粉中有效成分葫芦素B、葫芦素E的含量在≤8 kGy辐照后无明显变化(P> 0. 05),但经10 kGy辐照后,葫芦素B、葫芦素E含量变化显著(P <0. 05)。结论:辐照剂量为8 kGy时,~(60)Co-γ射线可作为天花粉药材辐照灭菌方法。
Objective: To study the efficacy of sterilization and explore the effect of ~(60)Co-γ ray irradiation on the effective components of Radix Trichosanthis. Methods: The sliced crude materials of Radix Trichosanthis were irradiated by ~(60)Co-γ ray respectively at 0,5,8 and 10 kGy. The changes in the total numbers of aerobic bacteria,molds and yeast before and after the sterilization were investigated by microbiological limit examination and the content changes of the active ingredients cucurbitacch B and cucurbitacch E were detected by HPLC. Results: After 8 kGy radiation,Radix Trichosanthis could meet the microbial limit requirements of Chinese Pharmacopoeia. There were no significant changes in the contents of cucurbitacch B and cucurbitacch E in Radix Trichosanthis after ≤8kGy irradiation (P > 0. 05),but there were significant changes in the contents of cucurbitacch B and cucurbitacch E after 10 kGy radiation (P < 0. 05). Conclusion:~(60)Co-γ ray at 8 kGy can be used as the dosage for irradiation sterilization of Radix Trichosanthis.
