响应面法优化左归饮总多糖提取工艺研究

摘要
目的:应用响应面法对左归饮中总多糖成分的提取工艺进行优化研究。方法:采用水提醇沉法对左归饮总多糖进行提取,苯酚-硫酸法测定总多糖含量,在单因素实验基础上,以总多糖含量为响应值,通过四因素三水平Box-Behnken设计实验,利用响应面法(RSM)分析确定左归饮总多糖的最佳提取工艺。结果:最佳工艺条件为料液比1:10.61,提取温度81.31℃,提取时间44.72min,醇沉后乙醇体积64.37%。结论:本实验所采用的方法操作简便,提高了总多糖提取效率,为左归饮总多糖的进一步研究奠定基础。
        Objective: To optimize the extraction process of polysaccharide in Zuoguiyin by using the response surface method.Method: The polysaccharide in Zuoguiyin was extracted by water extraction and alcohol precipitation and determined content by phonel-sulfate method. Based on single factor experiments,response value for polysaccharide content,four factors of the three levels of the Box-Behnken Design was established,and response surface method( RSM) analysis was used to determine the best extraction processof the polysaccharide in Zuoguiyin. Result: The optimal parameters were as follows,the solid-liquid ratio was 1: 10.61,extraction temperature was 81.31℃,extraction time was 44.72 min and alcohol sedimentation volume was 64.37%. Conclusion: The method is simple and easy to operate,and will lay the foundation for the further research of the polysaccharide in Zuoguiyin.
