摘要
目的:对山银花提取物进行定性定量研究,为其质量控制提供依据。方法:采用Acclaim 120 C18色谱柱(250 mm×4.6 mm,5μm),乙腈-0.4%磷酸溶液为流动相进行梯度洗脱,流速为1.0 ml·min-1,检测波长327 nm,柱温为25℃,测定绿原酸的含量;并以绿原酸为参照物,通过特征图谱鉴别6种有机酸。同时采用HPLC法对灰毡毛忍冬皂苷乙进行定性鉴别。结果:绿原酸在23.61~141.65μg·ml-1范围内线性关系良好(r=0.999 8),回收率为98.8%,RSD 1.4%(n=6)。灰毡毛忍冬皂苷乙为山银花提取物中的特征性成分,专属性强。结论:本法简便、可靠、准确,可用于山银花提取物的质量控制。
Objective: To establish qualitative and quantitative methods for Lonicerae Flos extract to provide basis for the quality control. Methods: An Acclaim 120 C18 column( 250 mm × 4. 6 mm,5 μm) was used at the temperature of 25℃. The mobile phase consisted of acetonitril and 0. 4% phosphoric acid solution with gradient elution. The flow rate was 1. 0 ml·min-1,the detection wavelength was 327 nm and the column temperature was 25℃. Chlorogenic acid was determined quantitatively and 6 organic acids were detected qualitatively. Dipsacoside B was identified by HPLC as well. Results: Chlorogenic acid had a good linear correlation within the range of 23. 61-141. 65 μg·ml-1( r = 0. 999 8). The average recovery was 98. 8%,and the RSD was 1. 4%( n = 6). Dipsacoside B was the exclusive content of Lonicerae Flos extract. Conclusion: The method is simple,reliable and accurate,and can be used for the quality control for Lonicerae Flos extract.
