脑苷肌肽对有机磷中毒性迟发性神经病变大鼠学习记忆能力及血浆钙激活的中性蛋白酶、神经疾病靶标酯酶影响研究

摘要
目的:探讨脑苷肌肽注射液对有机磷中毒性迟发性神经病变(OPIDN)大鼠学习记忆能力及血浆钙激活的中性蛋白酶(CANP)、神经疾病靶标酯酶(NTE)的影响。方法:先对健康雄性SD大鼠进行学习记忆功能筛选,选择其中20只大鼠作为空白组,而后对其余大鼠进行乐果乳油给药造模,乐果乳油给药14d后选择其中症状相似的60只完全随机分为三组:对照组[蒸馏水1.25ml/(kg·d)]、甲钴胺组[甲钴胺片1.25mg/(kg·d)]、联合组[甲钴胺片1.25mg/(kg·d)、脑苷肌肽注射液1.25ml/(kg·d)]。对不同分组的大鼠进行相应的治疗,测定治疗4周后大鼠学习记忆能力,分别测定治疗2、4周后大鼠血浆组织中NTE和CANP表达水平。结果:染毒大鼠干预前血浆NTE蛋白水平显著低于空白组大鼠(P<0.05),甲钴胺组和联合组大鼠治疗后血浆NTE水平上升(P<0.05),其中联合组大鼠治疗后血浆NTE水平最高(P<0.05)。染毒大鼠干预前血浆CANP蛋白水平显著高于空白组大鼠(P<0.05),甲钴胺组和联合组大鼠治疗后血浆CANP水平下降(P<0.05),其中联合组大鼠治疗后血浆CANP水平最低(P<0.05)。染毒后的三组大鼠中,联合组大鼠记忆再现能力最佳(P<0.05)。结论:脑苷肌肽注射液对于OPIDN大鼠的记忆能力和神经细胞损伤修复均具有积极作用。
        Objective:To investigate the effects of enantioside injection on learning and memory ability and plasma CANP and NTE in OPIDN rats.Methods:The healthy male Sprague-Dawley rats were screened for learning and memory.Twenty rats were selected as blank group(group B),and the remaining rats were treated with dimethoate EC.After 14 days of administration of dimethoate,60 rats with similar symptoms were randomly divided into3 groups:control group(group C),mecobalamin group(group M)and combination therapy group(group N).Different groups of rats were treated accordingly.The learning and memory abilities of the rats were measured after four weeks of treatment.The levels of neurological disease target esterase(NTE)and calcium-activated neutral protease(CANP)in plasma tissue were measured before and 2 weeks and 4 weeks after treatment.Results:The level of plasma NTE protein in the poisoned rats before treatment was significantly lower than that in the blank group(P<0.05).The levels of plasma NTE in group M and group N increased after the treatment(P <0.05).The levels of plasma NTE were the highest in group N after the treatment(P<0.05).The level of plasma CANP protein in thepoisoned rats before treatment was significantly higher than that in the blank group(P<0.05).The levels of plasma CANP in group M and group N decreased after the treatment(P<0.05).The levels of plasma CANP were the lowest in group N after the treatment(P<0.05).In the three groups after poisoning,the memory ability of the N group was the best(P<0.05).Conclusion:Intracerebral enantioside injection has a positive effect on the memory ability and repair of nerve cell injury in OPIDN rats.
